摘要
目的:研究秦皮甲素(esculin,ES)对角叉菜胶致小鼠足肿胀及炎症反应的作用及其机制。方法:以角叉菜胶诱发小鼠足跖炎症模型为实验动物模型。末次给药后,热板法检测痛阈值,并检测小鼠足肿胀;取足跖组织制备匀浆液,ELISA法测定足跖组织中炎症因子IL-1β和TNFα水平。结果:与正常对照组比较,角叉菜胶模型组小鼠足肿胀度显著增高(P<0.01),足跖组织中IL-1β和TNFα含量显著增加(P<0.01,P<0.01)。与角叉菜胶模型组比较,ES可显著提高痛阈(P<0.05);ES可显著降低角叉菜胶诱导的足肿胀及炎症因子IL-1β和TNFα水平(P<0.01,P<0.05,P<0.05)。结论:ES对角叉菜胶致小鼠足肿胀具有显著改善作用,机制可能与其抑制炎症因子合成及释放相关。
Objective: To observe the effects of esculin( ES) on the swelling of toes induced by carrageenan and explore its anti-inflammatory mechanism in mice. Methods: The experimental model of toes swelling in mice was induced by carrageenan given subcutaneously. The pain threshold was tested by hot plate test. The swelling of mouse toes was measured by vernier caliper,and the levels of IL-1β and TNFα were determined by ELISA assay.Results: Compared with the control group,the swelling of toes and the levels of IL-1β and TNFα were all significantly increased in mice injected with carrageenan( all P〈0. 01). Compared with the model group,esculin could significantly increase the pain threshold of mice( P〈0. 05),inhibit the carrageenan-induced swelling of toes( P〈0. 01) and decrease the levels of IL-1β and TNFα( both P〈0. 05). Conclusion: Esculin can significantly increase the pain threshold and inhibit the swelling of toes induced by carrageenan in mice. The mechanism may be attributed to its inhibition of inflammatory factors.
