龙胆多糖的不同提取工艺及抗氧化活性研究 被引量:6

STUDY ON DIFFERENT EXTRACTION TECHNOLOGY OF POLYSACCHARIDES FROM GENTIANA SCABRA AND ITS ANTIOXIDATION ACTIVITY

摘要 研究回流法、超声波辅助法和微波辅助法提取龙胆多糖的工艺,并考察龙胆多糖体外抗氧化活性。以多糖提取率为研究指标,在单因素试验的基础上,采用正交和响应面优化试验分别对回流法、超声法和微波法的提取工艺进行优化。结果表明:微波辅助萃取技术明显优于超声波法和回流法,其最佳工艺条件为提取温度87℃、料液比1∶20(g/m L)、提取时间18.93 min、微波功率900 W,在此条件下,龙胆多糖的提取率可达到13.28%。对龙胆多糖的体外抗氧化活性进行研究,结果表明龙胆多糖具有一定的还原力和羟基自由基清除能力。 Three methods including heat reflux, ultrasotmd-assisted and microwave-assisted were employed to extract polysaccharide from Gentiana scabra in this study, and its in vitro antioxidant activity was also studied with vitamin C as a reference. Taking the extraction efficiency of polysaccharides as an index, orthogonal tests and response surface methodology were used to optimize the extraction conditions of heat reflux, ultrasound-assisted method and microwave-assisted method based on single factors experiment including extraction temperature, ratio of solid and liquid, and extraction time. Results showed that microwave-assisted method was superior to heat reflux and ultrasound-assisted methods, and its detailed optimum conditions were as follows: extraction temperature was 87 ℃, ratio of solid and liquid was 1:15, extraction time was 18.93 min and microwave power of 900 W. Under the optimized extraction conditions, the extraction efficiency of polysaccharides could reach 13.28%. The antioxidant activity of polysaccharide was also evaluated, results showed that polysaccharides possessed significant antioxidant activity in vitro, the reducing power and scavenging ability against hydroxyl radicals were enhanced with increasing of polysaccharides concentration. The results also indicated that scavenging ability against hydroxyl radicals of polysaccharide was higher than that of Vc when the concentration was lower than 4.5 g/L.
