摘要
目的 描述四川省急性弛缓性麻痹(acute flaccid paralysis,AFP)病例中埃可病毒6型(echovirus 6,E6)的基因特征.方法 对四川省2007-2014年AFP病例中分离到的11株E6进行全VP1区逆转录-聚合酶链反应(RT-PCR)扩增和核酸序列测定,进行序列比对和构建进化树.结果 从2889份AFP病例的粪便标本中分离出11株E6病毒.经测序鉴定为C2基因亚型10株,C4基因亚型1株.来源病例的临床诊断分别为:肠道病毒感染6例,周围神经炎l例,格林巴利综合征2例,肌炎2例.C4亚型与10株C2亚型VP1区核酸序列同源性为83.9%-85.2%,氨基酸同源性为96.5%-97.2%;10株C2亚型VP1区核酸序列同源性为94.0%-100.0%,氨基酸同源性为98.9%-100%.结论 2007-2014年四川地区AFP病例中分离到的E6为C2和C4基因亚型,其中以C2基因亚型为主.
Objective To describe the genetic characteristics of echovirus 6 (E6) among acute flaccid paralysis (AFP) cases in Sichuan province.Methods From 2007 to 2014,E6 strains from AFP cases in Sichuan province were isolated and the complete VP1 genes were amplified by the reverse transcriptionpolymerase chain reaction (RT-PCR).The PCR products were sequenced directly,then these sequences were aligned and the phylogenetic tree was constructed.Results Eleven E6 strains were isolated among 2889 fecal specimens,among which 10 strain were identified as subgenotype C4 and the other one as subgenotype C2.These strains were isolated from 6 cases of Enterovirus infection,2 cases of Guillain-Barre syndrome,2 cases of myositis,and one case of peripheral neuritis.Between the one C4 strain and 10 C2 strains,the nucleotide identity and the amino acid identity were 83.9%-85.2% and 96.5%-97.2%,respectively.The nucleotide identity and the amino acid identity among 10 C2 strains were 94.0%-100.0% and 98.9%-100%,respectively.Conclusions The E6 strains from AFP cases were identified as subgenotype C2 and C4 in Sichuan province from 2007 to 2014,and the most common subgenotype of E6 was C2.
