薯蓣总皂苷水解物中提取薯蓣皂苷元工艺研究

摘要 目的:优选一种能高效、安全地从薯蓣总皂苷水解物中提取薯蓣皂苷元的工艺。方法:以薯蓣皂苷元得率为指标,通过单因素试验对回流提取法、索氏提取法、超声提取法3种提取方法以及甲醇、乙醇、乙酸乙酯、石油醚、环己烷5种提取溶剂进行考察,采用正交试验法优化所选提取方法,确定最佳提取条件。结果:以石油醚为溶剂,采用超声提取法,最佳提取条件为:50倍料液比,频率40 k Hz,每次提取20 min,共3次。结论:超声法提取具有工艺周期短、效率高、能耗低、安全等优点,适合用于薯蓣皂苷元提取。 Objective: To optimize an efficient and safe method for the extraction of diosgenin from the total dioscin hydrolysate. Methods: Evaluated by the yield of diosgenin, three methods including refluxing extraction, soxhlet extraction and ultrasonic extraction with 5 extraction solvents including methanol, ethanol, ethyl acetate, petroleum ether and cyclohexane were optimized by single factor experiments. The orthogonal experiment method was used to optimize the extraction conditions of the selected method. Results: The ultrasonic extraction was selected, and petroleum ether was used as the extraction solvent. The best extraction conditions were as follows: the ratio of solid to liquid was 1:50, the ultrasonic frequency was 40 kHZ, extraction time was 20min, and extracting 3 times. Conclusion : The ultrasonic extraction process has the advantages of short cycle, high efficiency, low energy consumption and promising safety, which is suitable for the diosgenin extraction.
