额尔齐斯河鱼类宽头鲤蠢绦虫的种类鉴定及流行特点分析

Species identification and analysis of epidemic characteristics of Caryophyllaeus laticeps in fishes in the Irtysh River

摘要 为了研究额尔齐斯河鱼类肠道寄生的宽头鲤蠢绦虫(Caryophyllaeus laticeps)种类及流行特点,试验采用形态学和分子生物学相结合的方法鉴定虫种,并对额尔齐斯河鱼类8次采样获得的数据进行统计分析。结果表明:依靠形态学观察可知虫体不分节,头节宽大,体末端有"H"状卵巢,初步鉴定为鲤蠢绦虫属种类;通过扩增线粒体细胞色素c氧化酶第1亚基(cox1)序列,发现该虫与Gen Bank上公布的宽头鲤蠢绦虫的线粒体cox1序列的相似性为97.56%(639/655),为同一种。被宽头鲤蠢绦虫感染的5种鱼中,散鳞镜鲤的感染率最高(为16.67%),感染强度最低(为1);东方欧鳊的感染率、感染强度均高,分别为15.27%和14;银鲫的感染率最低,为3.28%。说明本试验采用的方法能够准确鉴定虫种;东方欧鳊感染宽头鲤蠢绦虫的情况最严重,且体长段适中的东方欧鳊更易感染宽头鲤蠢绦虫。 To study the species and epidemic characteristics of Caryophyllaeus laticeps parasitizing in the intestines of fishes in the Irtysh River. The combination methods of morphology and molecular biology were used to identify the species, and the data that obtained eight times from the fishes in the Irtysh River by sampling were used for statistical analysis. The result showed that relying on morphological observation, the body was unsegmented with wide scolex and "H" shaped ovaries in the ends of the body, the parasite was preliminarily identified as Caryophyllaeus. It was found that by amplifying mitochondrial cytochrome c oxidase subunit 1 ( coxl ) sequence, the coxl sequence shared 97.56% (639/655) with that of Caryophyllaeus laticeps published in GenBank, and the parasite belonged to the same species. Cyprinus carpio, var. specularis amu- rensis had the highest infection rate of 16.67% and the lowest infection intensity of 1 among five fishes infected by Caryophyllaeus laticeps; Abramis brama orientalis had higher Infection rate of 15.27% and infection intensity of 14; Carassius auratus gibelio had the lowest infection rate of 3.28 %. The results indicate that the combination methods used in the test can accurately identify parasite species; Abramis brama orien- talis are particularly prone to infect, it was found that medium length were more likely to be infected with Garyophyllaeus laticeps.
