摘要
目的探讨巴戟天萃取物对环磷酰胺(CTX)诱导生精障碍大鼠睾丸支持细胞的影响。方法 48只成年雄性SD大鼠,随机分为6组,空白对照组、CTX+NS组、CTX+MO 10 g/kg组、CTX+MO 20 g/kg组、CTX+MO 30 g/kg组、CTX+MO 40 g/kg组。以CTX构建大鼠生精障碍动物模型;巴戟天水提物用乙酸乙酯萃取后取水层成分,分别以10、20、30、40 g/(kg·d)的浓度给予4个剂量实验组灌胃,空白对照组及CTX+NS组予生理盐水灌胃,持续2周。取各组大鼠睾丸组织,荧光定量PCR检测卵泡刺激素受体(FSHR)mRNA和雄激素结合蛋白(ABP)mRNA表达的差异。结果 4个剂量实验组FSHR mRNA和ABP mRNA表达明显高于CTX+NS组及空白对照组,差异有统计学意义(P<0.05)。结论巴戟天萃取物对CTX诱导生精障碍大鼠的睾丸支持细胞具有保护作用,进而修复睾丸的生精功能。
Objective To study the impact of Morinda Officinalis (MO) extract on sertoli cells of adult male rats with cytoxan (CTX) - induced impaired Spermatogenesis. Methods We randomly divided 48 adult SD male rats into 6 groups : blank control group, CTX + NS group, CTX + MO10 g/kg group, CTX + MO 20 g/kg group, CTX + MO 30 g/kg group, and CTX + MO 40 g/kg group. The models of impaired testis in the SD rats by established by intraperitoneal injec- tion of CTX. The subjects in treatment group were treated with MO at the concentrations of 10, 20,30, and 40 g/kg, re- spectively. After two weeks of medication, the expression of FSHR mRNA and ABP mRNA was compared with real - time fluorescence quantitative PCR. Results The expression of FSHR mRNA and ABP mRNA in the four treatment groups were significantly higher than CTX + NS group. Conclusion MO extract can protect the sertoli cells of adult male rats from Cytoxan- induced impaired Spermatogenesis.
