血浆miRNA-24的异常表达与冠状动脉病变程度的相关性 被引量:5

摘要 目的探讨不同冠状动脉病变程度患者的血浆miRNA-24表达水平的差异,研究miRNA-24对冠脉病变患者的临床意义。方法入选在心内科就诊的,已行冠状动脉造影的患者97例,其中对照组25例,冠心病组72例,根据症状、心电图、心肌酶、造影结果将冠心病组分为稳定型心绞痛组(28例),不稳定型心绞痛组(21例)和急性心肌梗死组(23例)3个亚组。采集各组一般临床资料,抽血检测相关血液及生化指标,荧光实时定量PCR方法检测miRNA-24水平并记录Gensini评分,分析miRNA-24在各组中的差异表达,及研究miRNA-24表达水平与Gensini评分的相关性。结果冠心病患者各分组(稳定型心绞痛、不稳定型心绞痛及急性心肌梗死组)miRNA-24均低于对照组,差异有统计学意义(P<0.05);其中不稳定型心绞痛组与急性心肌梗死组miRNA-24低于稳定型心绞痛组,差异均有统计学意义(P<0.05);不稳定型心绞痛组与心肌梗死组差异无统计学意义(P>0.05)。冠心病组Gensini评分与miRNA-24表达水平呈负相关。结论冠心病患者miRNA-24表达水平降低,且与Gensini评分呈负相关,其高表达对冠状动脉有一定保护作用。 Objective To investigate the differences in plasma miRNA -24 expression levels in patients with different degree of coronary artery lesions and so as to study the clinical significance of miRNA - 24 in patients with coronary artery disease (CHD). Methods A total of 97 patients, who received coronary angiography in Cardiology Department of our hospital, were selected, including 25 cases in the control group and 72 cases in CHD group. According to the symptoms, ECG, cardiac enzymes and the finding in gangiography, the CHD group were subdivided into three different subgroups, stable angina subgroup (28 cases, Subgroup SAP), unstable angina (21 cases, Subgroup UAP) and acute myocardial infarction (23 cases, Subgroup AMI). In each groups, general clinical data, blood biochemical report, as well as Gensini score were recorded. The expression of miRNA - 24 was tested by rt - PCR. By analyzing the difference of miRNA ' 24 expression levels in each group, the correlation with miRNA - 24 expression levels and Gensini score was studied. Results The patients in 3 subgroups of CHD had significantly lower miRNA -24 expression levels than the control group (P 〈 0. 05 ). The miRNA - 24 expression levels of the patients in UAP subgroup and AMI subgroup were significantly lower than SAP group (P 〈 0. 05 ). Between UAP group and AMI group, there was no significant difference in miRNA - 24 expression ( P 〉 0. 05 ). In CHD group, Gensini score and miRNA - 24 expression was negatively correlated. Conclusion The expression level of miRNA -24 decreases in patients with CHD, which is negatively correlated with C, ensini score. High miRNA -24 expression level has some protective effect on coronary artery.
