摘要
目的研究活血通络汤治疗家兔激素性股骨头缺血性坏死的作用机制。方法将40只日本大耳兔随机分为空白组、模型组、治疗给药组、预防加治疗给药组,每组10只,采用贺西京法制做家兔激素性股骨头坏死模型。预防给药组第2周开始喂服中药饲料,治疗给药组第3周开始喂服中药饲料,模型组和空白组均喂服等量普通饲料。第8周末各实验组耳缘静脉取血,RT-PCR检测白细胞Notch1,Notch2,Notch3,Notch4、DLL1,DLL3,DLL4,Jagged1,Jagged2,Hes1,Hes5,HERP1,HERP2基因表达。同时对股骨头进行病理镜检,计算空骨陷窝率。结果第8周时,活血通络汤预防给药组和治疗给药组家兔股骨空骨陷窝率显著低于模型组(P<0.05);相比模型组,预防给药组家兔白细胞Notch1和Notch3 mRNA表达有显著升高(P<0.05),而Notch2和Notch4 mRNA表达有显著下调(P<0.05);相比模型组,预防给药组和治疗给药组家兔白细胞Hes1和Hes5 mRNA表达有显著下调(P<0.05),Jagged1 mRNA表达有显著升高(P<0.05);预防给药组和治疗给药组家兔白细胞DLL1和DLL4mRNA表达有显著升高(P<0.05)。结论活血通络汤通过早期作用于Notch1或其它成员导致其表达发生变化,从而激活Notch如Notch1-DLL1信号传导通路,而增加Notch1胞内NICD的表达,促进下游靶基因的转录表达,促进血管生成,从而参与对骨细胞增殖的调控,进而促进新骨生成。
Objective To study the cure mechanism of HuoxueTongluo Deeoetion on cure of induced avascular necrosis of femoral head based on Notch signal pathway. Methods 40 rabbits were divided into blank group, model group, treatment group and pre- vention + treatment group with 10 rabbits in each group. Model animal were achieved by He's method. Prevention + treatment group were given traditional herb since 2ed week. Treatment group were given traditional herb sinee 3rd week. Blank and model group were given basic feed as same amounts as treatment group. Venous blood sampled for the measuring of expression of Notchl, Notch2, Notch3, Notch4, DLL1, DLL3, DLIA, Jagged1, Jagged2, Hes1, Hes5, HERP1 and HERP2. Percentage of empty lacunae were calculated by microscopic pathology examination of femoral head. Results Percentage of empty lacunae of treatment group were significantly lower than that of model group( P 〈 0.05 ). Expression of Notch 1 , Notch3, Dell1 , Dell4, J aggedl in prevention + treatment group were significantly than that in model group( P 〈 0. 05) , but N oteh2, Notch4, Hesl and Hess showed an opposite results. Conclusion The pathway of Notch1 - DLL1 were activated by HuoxueTongluo Deeoction's affection on Notehl to inerease the expression of NICD, thus promote genesis of hones.
