摘要
目的:评价大黄有效成分大黄素对急性肺损伤(ALI)的保护作用,探讨其防治急性肺损伤的作用机制。方法 SD大鼠40只随机分为四组:空白对照组、模型组、溶剂组、大黄素组,每组各10只。采用气管内滴注内毒素法制备ALI大鼠模型,溶剂组腹腔注射脂肪乳,大黄素组腹腔注射的剂量为10mg/kg,在造模前24h,造模,造模后24h内行腹腔注射给药,空白对照组不作任何处理。在造模后48h采集标本,并且行肺组织HE染色,进行肺泡灌洗液中总细胞数及中性粒细胞计数,检测其总蛋白含量及乳酸脱氢酶,采用酶联免疫法检测大鼠BALF中TNF-α、IL-1β的含量。结果相比较与模型组与溶剂组,大黄素组能显著减轻肺间质炎性细胞浸润,肺间质水肿,肺泡壁增厚,肺泡腔破坏程度。且可以降低肺泡灌洗液中的总细胞数和TNF-α、IL-1β(P〈0.05),但其对中心粒细胞比值及总蛋白含量影响不大(P>0.05);另外其可以引起BALF中的LDH升高(P〈0.05)。结论大黄素对急性肺损伤存在一定的治疗作用,这可能与其降低肺组织中TNF-α、IL-1β等促炎因子,从而降低肺泡灌洗液中的总细胞数有关,但其可以引起肺泡灌洗液LDH的升高。
Objective In this study,to explore the rapeutic effect of the Angelica monomer “Emodin”in acute lung injury by animal experiments. Methods 40 SD rats were randomly divided into seven groups:control group,model group,solvent group,and Emodin groups.Each group have 10 rats.ALI rat model prepared by Using intratracheal instillation of LPS,the solvent group by intraperitoneal injection of fat emulsion,the monomer groups by intraperitoneal injection of dose 10mg/kg in 24 hours before modeling,modeling,and 24 hours after modeling. Nothing was given in blank control group.Samples were collected 48 hours after modeling,then do lung tissue H&E staining,count the total number of cells and the rato of neutrophil in the bronchoalveolar lavage fluid. Detect of the total protein content and lactate dehydrogenase,the enzyme-linked immunosorbent assay in BALF and various tissues of TNF-α,IL-1βcontent in BALF are also assay by ELISA(enzyme-linked immunosorbent assay). Results Compared with the model group and the solvent group,Emodin significantly reduced the infiltration of pulmonary interstitial inflammatory cell,edema of interstitial pulmonary,also the thickening of alveolar wall,the alveolar damage. and the total number of cells,TNF-α,IL-1βin bronchoalveolar lavage fluid(P〈0.05).But neutrophil ratio and total protein content not affected(P>0.05);In addition,it can cause the BALF LDH increased(P〈0.05). Conclusion The therapeutic effect of Emodin in acute lung injury may relate its decreasing important pro-inflammatory cytokines in lung tissue like TNF-αand IL-1βwhich reduce the total number of cells in BALF. But it can cause elevated LDH in BALF.
