基于聚类分析及主成分分析的丹莪妇康煎膏的HPLC指纹图谱 被引量:3

HPLC Fingerprint of Dan'e Fukang Decocted Extract Based on Hierarchical Cluster Analysis Coupled with Principal Component Analysis

摘要 建立了丹莪妇康煎膏的HPLC指纹图谱。采用Welch Ultimate XB—C18色谱柱,流动相为乙腈:0.1%甲酸,梯度洗脱,检测波长230nm(0~60min)和254nm(60~110min)。将10批丹莪妇康煎膏的HPLC指纹图谱导入《中药色谱指纹图谱相似度评价系统》2004A版,对共有峰进行聚类分析及主成分分析,并通过比对对照品和药材的色谱保留时间与uV特征吸收,对主要特征峰进行归属指认。结果,10批丹莪妇康煎膏的指纹图谱相似度均大于0.98。 An HPLC fingerprint was established for the quality control of Dan'e Fukang decocted extract (DFDE). A Welch Ultimate XB-CI8 column was used, with the mobile phase of acetonitrile and 0.1% formic acid by gradient elution, at the detection wavelength of 230 nm (0-60 min) and 254 nm (60-110 min). The HPLC fingerprint spectra for 10 batches of DFDE were obtained and evaluated by Similarity Evaluation System for Chromatographic Fingerprint of TCM (2004A edition). Further discussions were made on the basis of hierarchical cluster analysis (HCA) and principal component analysis (PCA) for common peaks. Furthermore, the main characteristic peaks of DFDE and herbal materials were identified by comparing their retention time and the UV characteristic absorption with those of the chemical references. The similarities of 10 batches of DFDE were all above 0.98.
