摘要
以合浦珠母贝珍珠层粉为原料,采用水提法提取珍珠层粉的水溶性基质,提取率为0.212%。采用分子截留量为8 k Da与1 k Da的透析膜对水溶性基质进行透析,得到3种亚组分:WSM>8 000,1 000<WSM<8 000和WSM<1 000,提取率分别为17%,7.4%和33.5%。采用蘑菇酪氨酸酶多巴速率氧化法测定3种亚组分对酪氨酸酶活力的抑制作用,其中组分WSM>8 000对酪氨酸酶活力具有最强的抑制作用,IC_(50)为8.80 mg/m L,对其进行SDS-PAGE电泳分析发现样品中存在一种未曾见文献报道的蛋白条带;组分WSM<1 000的抑制作用次之,IC_(50)为27.3 mg/m L;组分1 000<WSM<8 000对酪氨酸酶活力具有一定的抑制作用,但其作用质量浓度与酪氨酸酶活力抑制率不呈剂量依赖关系。
The water soluble matrix from Pinctadafucata nacre powder was extracted using water, representing 0.212% of the nacre weight. Dialysis of the matrix through 8 kDa and 1 kDa cut-off membranes showed that 17% of it was made of protein and polypeptide which was named WSM〉 8 000, 33.5% of it was made of small molecules of molecular masses below 1 kDa which was named WSM〈1 000, and the remaining 7.4% was named 1 000〈WSM〈8 000. Their inhibitory effects on mushroom tyrosinase were evaluated, and the results indicated that compound WSM〉 8 000 had stronger inhibition of tymsinase activity than compound WSM〈 1 000 and compound 1 000〈WSM〈 8 000. The IC50 of compound WSM〉 8 000 and WSM〈 1 000 were 8.80 mg/mL and 27.3 mg/mL. Compound 1 000〈WSM〈8 000 had certain inhibitory effect on tyrosinase activity but no dose-response relationship with it.
