摘要
目的探讨卵巢癌腹水及外周血CD4^+CD25^+Treg/CD4^+T细胞含量及意义。方法选择确诊为卵巢癌合并腹水的患者40例为研究对象,采用流式细胞术、ELISA法检测腹水及外周血中CD4^+CD25^+Treg/CD4^+T、血清中肿瘤标志物、腹水中基质蛋白酶含量,并与正常对照组进行比较。结果卵巢癌患者腹水中CD4^+CD25^+Treg/CD4^+T细胞含量(28.44±3.12)%明显高于外周血中CD4^+CD25^+Treg/CD4^+T细胞含量(14.72±2.14)%(P<0.05);浆液腺癌腹水、外周血中CD4^+CD25^+Treg/CD4^+T细胞含量明显高于黏液腺与其他组织学类型,Ⅲ~Ⅳ期明显高于Ⅰ~Ⅱ期,G1分化程度明显高于G2、G3分化程度(P<0.05)。卵巢癌组癌抗原125(CA125)、人附睾蛋白4(HE4)、血管内皮生长因子(VEGF)明显高于正常对照组(P<0.05),卵巢癌组患者腹水中MMP2、MMP9含量明显高于正常对照组(P<0.05)。结论卵巢癌腹水中CD4^+CD25^+Treg/CD4^+T细胞含量明显高于血清中CD4^+CD25^+Treg/CD4^+T细胞含量,不同组织学类型、肿瘤分期、分化程度表达各不相同,且与肿瘤标志物含量、MMPs含量升高呈正相关。
Objective To explore CD4+CD25+ Treg/CD4+T cells contents and significance in ascites andperipheral blood of ovarian cancer patients. Methods Forty patients with ovarian cancer combined with aseites were selected, flow cytometry and ELISA were used to detect the contents of CD4+CD25+ Treg/CD4+T cells in ascites and peripheral blood, serum tumor markers, and matrix metalloproteinase (MMP) in ascites, then the results were compared with normal control group. Results The content of CD4+CD25+ Treg/CD4+T cells in ascites of ovarian cancer patients was (28.44±3.12) %, which was higher than that in peripheral blood [ ( 14. 72±2. 14) % ] ( P〈0. 05 ) ; the content of CD4+CD25+Treg/CD4+T cells in ascites and peripheral blood of patients with serous adenocarcinoma were higher than those of patients with other histological types (P〈0. 05 ) ; the contents of CD4+CD25+ Treg/CD4+T cells in ascites and peripheral blood of patients with ovarian cancer of stage Ⅲ-Ⅳ were higher than those of patients with ovarian cancer of stage Ⅰ-Ⅱ ( P〈0. 05 ) ; the contents of CD4+CD25+ Treg/CD4+T ceils in ascites and peripheral blood of patients with ovarian cancer of G1 were significantly higher than those of patients with ovarian cancer of G2, G3. The levels of CA125, HE4, VEGF in peripheral blood and MMP2 and MMP9 in ascites in ovarian cancer group were higher than those in normal control group (P〈0. 05) . Conclusion The content of CD4+CD25+ Treg/CD4+T cells in ascites of ovarian cancer patients is higher than that in serum, ther are differences in ovarian cancer of different histological types, tumor stages, and differentiation degrees, which is positively correlated with increases of tumor markers and MMPs contents.
