摘要
目的探讨血府逐瘀胶囊在模拟失重状态下对骨骼肌萎缩的防护作用。方法建立尾部悬吊大鼠模拟失重模型,30只同源同窝雄性SD大鼠随机分为对照组、尾吊组和血府逐瘀组(尾吊+血府逐瘀胶囊),每组10只。对照组正常饲养,尾吊组和血府逐瘀组均尾吊30 d。对照组及尾吊组单给予纯溶媒,血府逐瘀组给予血府逐瘀胶囊,剂量为25 mg/(kg·d),每组8:00灌胃,1次/d。(1)测量比目鱼肌湿重体质量比和横截面积。(2)应用免疫蛋白质印迹法(Western-blot)技术检测PI3K/AKT通路相关蛋白及下游分子GRP78、BAX、Cyto-c蛋白表达水平。(3)应用caspase-3活性检测试剂盒检测caspase-3活性。结果 30只大鼠全部进入结果分析,无脱失。(1)与对照组相比,尾吊组比目鱼肌平均湿重体质量比下降了56.39%;血府逐瘀组平均湿重体质量比较单纯尾吊组增加了14.49%,且差异均有统计学意义(P<0.05)。(2)尾吊30 d显著抑制了PI3K/AKT信号通路中PI3K、p-AKT的蛋白表达水平,与对照组比较,血府逐瘀组干预后提高了PI3K、p-AKT的表达,其中对关键蛋白p-AKT的表达影响最大,与尾吊组比较,差异有统计学意义(P<0.05)。(3)尾吊30 d比目鱼肌caspase-3酶活性显著增强,血府逐瘀组caspase-3酶活性较尾吊组显著降低,差异有统计学意义(P<0.05)。结论 (1)尾吊模拟失重可通过抑制PI3K/AKT通路相关蛋白表达水平,增强caspase-3酶活性,导致比目鱼肌萎缩;(2)血府逐瘀胶囊可能通过提高PI3K/AKT通路相关蛋白表达水平,对失重性肌萎缩起到保护作用。
Objective To discuss the protective effect of Xuefu Zhuyu capsule on skeletal muscle atrophy under simulated weightlessness. Methods Tail-suspended rat was taken as the animal model of simulated microgravity. 30 homologous Sprague-Dawley male rats(aged 10 weeks, weighing 200-250g) were randomly divided into three groups, including control group, tail-suspended group, Xuefuzhuyu group(tail-suspended + Xuefu Zhuyu capsule). The 25 mg/(kg·d) Xuefu Zhuyu was administrated to the Xuefu Zhuyu group, while the 1ml/d solvent to the control group and tail-suspended group. All the rats received the intragastric administration at eight o'clock every day.(1) The soleus morphological changes were measured, including the wet weight of body weight, the cross-sectional area of muscle fibers.(2) The protein and m RNA expression of PI3K/AKT, and some downstream proteins such as BAX and Cyto-c were detected by Western-blot and RT-PCR.(3) The caspase-3 activity was detected by caspase-3 fluorometric assay kit. Results 30 rats were all taken in the result analysis, without any loss.(1) Compared with the control group, the wet weight of soleus muscle of the tail-suspended group decreased by 56.39%; compared with the tail-suspended group, the average wet weight of Xuefu Zhuyu group increased 14.49%, and the differences were both significant(P〈0.05).(2) After tail-suspended for 30 days, the content of P-AKT and GRP78 decreased under simulated microgravity, but the activity of Akt and GRP78 of Xuefu Zhuyu group increased compared with that of control group, and the differences was significant(P〈0.05).(3) The caspase-3 activity of soleus muscle increased after tail-suspended for 30 days. And the caspase-3 activity of Xuefu Zhuyu group was lower than that of tail-suspended group, and the differences was significant(P〈0.05). Conclusions(1) Simulated microgravity may play an important role in the soleus muscle atrophy, which increases the caspase-3 activity through inhibiting protein expression level related to the PI3K/AKT pathway.(2) Xuefu Zhuyu capsule may have protective effect on muscle atrophy induced by weightlessness by increasing protein expression level related to the PI3K/AKT pathway.
