Expression of FXYD6 in Hilar Cholangiocarcinoma Tissue and Its Clinical Significance

摘要 目的 检测FXYD6蛋白在肝门胆管癌及相应癌旁远端正常胆管组织中的表达并分析其临床意义。方法 应用免疫组织化学链酶亲和素-生物素-过氧化物酶复合物(SABC)法检测58例肝门胆管癌和30例远端正常胆管组织中FXYD6蛋白的表达,分析FXYD6蛋白表达与肝门胆管癌患者临床病理特征的关系。结果 FXYD6蛋白在肝门胆管癌组织中的表达阳性率明显高于其在癌旁远端正常肝门胆管组织中的表达阳性率,差异有统计学意义〔75.9%(44/58)比33.3%(10/30),χ^2=15.084,P=0.000〕。FXYD6蛋白在高、中分化肝门胆管癌组织中的表达阳性率明显高于其在低分化肝门胆管癌组织中的表达阳性率〔85.4%(35/41)比52.9%(9/17),χ^2=5.243,P=0.022〕,而FXYD6蛋白表达与肝门胆管癌患者的性别(χ^2=0.000,P=1.000)、年龄(χ^2=1.248,P=0.264)、T分期(χ^2=0.466,P=0.495)、淋巴结转移(χ^2=0.357,P=0.550)、病理分期(χ^2=0.005,P=0.944)及神经浸润(χ^2=3.016,P=0.082)均无关。结论 FXYD6在肝门胆管癌组织中的蛋白表达与其分化程度有关,其可能作为肝门胆管癌的一种新型肿瘤生物标志物。 Objective To detect expression of FXYD6 protein in hilar cholangiocarcinoma tissues and explore its significances. Methods The expressions of FXYD6 protein in the 58 hilar cholangiocarcinoma tissues and 30 normal bile duct tissues adjacent to cancer were detected by strept avidin-biotin complex (SABC) immunohistochemistry. The relation between FXYD6 protein expression and biological characteristics of patient with hilar cholangiocarcinoma was analyzed. Results The positive rate of FXYD6 protein expression in the hilar cholangiocarcinoma tissues was significantly higher than that in the normal bile duct tissues adjacent to cancer [75.9% (44/58) versus 33.3% (10/30), χ^2=15.084, P=0.000]. Furthermore, the positive rate of FXYD6 protein expression in the well and moderately differentiated hilar cholangiocarcinoma tissue was significantly higher than that in the poorly differentiated hilar cholangiocarcinoma [ 85.4% (35/41) versus 52.9% (9/17), χ^2=5.243, P=0.0221, which was not related to the gender (χ^2=0.000, P= 1.000), age (χ^2= 1.248, P=0.264), T stage (χ^2=0.466, P=0.495), lymph node metastasis (χ^2=0.357, P=0.550), pathological stage (χ^2=0.005, P=0.944), and perineural invasion (χ^2=3.016, P=0.082). Condustion The positive rate of FXYD6 protein expression is associated with differentiation of hilar cholangiocarcinoma, which might be a new biomarker of it.
