摘要
目的研究不同剂量X射线照射人三阴性乳腺癌细胞,照射后不同时间点对人三阴性乳腺癌细胞E-钙黏蛋白(E-cadherin)基因表达的影响。方法用X射线分2.5、5和10 Gy 3个吸收剂量组照射体外培养的人三阴性乳腺癌HCC1937细胞和MDA-MB-231细胞,采用荧光实时定量PCR技术检测HCC1937细胞和MDA-MB-231细胞经照射后6、12、24和48 h的E-cadherin mRNA表达水平,以未照射组(0 h组)为对照。结果 HCC1937细胞和MDA-MB-231细胞E-cadherin mRNA的表达在2.5、5、10 Gy X射线照射后,随着时间点的后移呈升高趋势。与未照射组相比,除了HCC1937细胞10 Gy照射后6 h、2.5 Gy和5 Gy照射后24 h及MDA-MB-231细胞5 Gy照射后12 h外,各时间点差异均有统计学意义(P<0.05)。HCC1937细胞在2.5 Gy和10 Gy照射后48 h,E-cadherin mRNA的表达呈下降趋势,与未照射组相比,差异有统计学意义(P<0.05)。结论较大剂量的X射线整体可上调HCC1937细胞和MDA-MB-231细胞E-cadherin mRNA的表达,可能诱导了肿瘤细胞凋亡;晚期HCC1937细胞E-cadherin mRNA的表达下降,可能与辐射所致HCC1937细胞的放射生物学效应有关。
Objective To investigate the effects of different doses of X-ray irradiation on the expression of E-cadherin gene in triple negative breast cancer ceUs. Methods Human triple negative breast cancer MDA-MB-231 and HCC1937 cells were exposed to X-ray Irradiation at a dose of 2.5, 5 and 10 Gy for 6, 12, 24 and 48 h, respectively. E-cadherin mRNA expression levels in HCC1937 and MDA-MB-231 cells after irradiation were detected with fluorescence real-time PCR technology. Results E-cadherin mRNA expression levels in HCC1937 and MDA-MB-231 ceils after irradiation were raised with exposure time. Except HCC1937 cell 6 h after 10 Gy, 2.5 Gy irradiation and 24 h after 5 Gy irradiation, MDA- MB-231 cells 12 h after 5 Gy irradiation, there were significant differences at all time points between irradiation and non-irradiation groups (P 〈 0. 05 ). E-cadherin mRNA expression of HCC1937 cells was decreased 48 h after 2.5 Gy and 10 Gy irradiation, compared with those of the non- irradiation group ( P 〈 0.05 ). Conclusion High dose of X-rays irradiation may upgrade the expression of E-cadherin mRNA in human HCC1937 and MDA-MB-231cells, resulting in cell apoptosis. The decreased expression of E-cadherin mRNA in HCC1937 cells after long exposure to X-ray irradiation might be associated with the biological effect of radiation.
