摘要
目的建立气相色谱内标法测定中链甘油三酯的含量。方法采用气相色谱内标法,以十六酸十六酯为内标物,100%聚二甲基硅氧烷为固定液(ZB-1)的非极性石英毛细管柱,柱温为290℃,进样口温度为300℃,火焰离子检测器温度为300℃,氮气流速为2.0 mL·min^(-1)进样量为1μL。结果辛酸甘油三酯在浓度范围为0.066~0.464 mg·mL^(-1)时,线性关系良好,线性方程为Y=3.872 3X+0.022 6,相关系数r^2=0.999 1,辛酸甘油三酯的定量限为0.03μg;癸酸甘油三酯在浓度范围为0.065~0.458 mg·mL^(-1)时,线性关系良好,线性方程为Y=4.185 6X-0.051 9,相关系数r^2=0.999 4,癸酸甘油三酯的定量限为0.06μg;方法的加样回收率为98.76%~101.0%。结论该方法简便、快速、准确,适用于测定中链甘油三酯主成分的含量。
OBJECTIVE To establish an internal standard method for Medium-chain triglyceride determination by GC. METHODS An internal standard substance was used by GC ( Palmityl palmitate). The separation was carried out on a ZB-1 capillary column with the oven temperature of 290 ℃. The temperature of the injection inlet was 300 ℃ and that of FID was 300 ~C. The nitrogen flow rate was 2.0 mL. min-1. Sample injection volume was 1 μL. RESULTS The linear range of Oetylic acid triglycerides was 0.066 - 0.464 mg·mL-1( Y= 3. 872 3X+0. 022 6, r2= 0.999 1 ),The quantitation limit of Octylic acid triglycerides was 0.03μg . The linear range of Decanoic acid triglycerides was 0.065-0.458 mg·mL-1( Y= 4.185 6X-0.051 9, r2 = 0.999 4), The quantitation limit of Decanoic acid triglyeerides was 0.06 μg. The recoveries of Medium-chain triglyceride were 98.76% - 101.0%. CONCLUSION The method of determination of Medium-chain triglyceride by GC is convenient, and accurate .
