正交法优选具有抗巴氏杆菌活性黄连的提取工艺

摘要 为了筛选出中药黄连有效部位的最佳提取工艺,以提取物有效部位的浸膏得率与对巴氏杆菌的最低抑菌浓度和最低杀菌浓度为综合考察指标,采用正交实验对提取工艺中的料液比、提取次数、提取时间、乙醇浓度4个因素进行优选研究。黄连有效部位的最佳提取方法为:70%乙醇,料液比1:12,提取次数2次,每次1 h。在该工艺条件下进行提取,中药有效部位浸膏得率可以得到最大化保障,对巴氏杆菌的MIC和MBC值均小于等于15.6mg/mL。 The aim of this study was to screen the best extraction procedure for anti Pastewella activity of Rhizomacoptidis effective parts.The extract yield of effective parts,minimal inhibitory concentration(MIC) of extracts on Pastewella and the minimum bactericidal concentration(MBC)of extacts on Pastewella were used as the three indicators for comprehensive evalu-ation.The optimized extraction procedure was screened by orthogonal design with four factors,the sohd-liquid ratio,extraction time,time of extractionand concentration of ethanol.Our result showed that the optimized concentration of ethanol was 70%,the solid-liquid ratio was 1:12,and the extraction time was 2 times of,1-hour extraction.In these conditions,Rhizoma coptidis effective parts are maximum,and MIC and MBC values for pastewella are less than or equal to 15.6 mg/mL.
