红景天苷联合胰岛素对高糖培养海马神经元凋亡及Bcl-2、Bax蛋白表达的影响 被引量:2

Effects of rhodioside combine with insulin on Bcl-2 and Bax expression and the apoptosis of hippocampal neurons cultured in high glucose

摘要 目的:探索红景天苷联合胰岛素对高糖培养海马神经元凋亡及Bcl-2、Bax蛋白表达的影响。方法:选择15只新生24h SD大鼠,取海马神经元进行原代培养,分为正常对照组(NC组)、高糖组(HG组)、胰岛素组(RI组)、红景天苷组(RH组)、胰岛素联合红景天苷组(RI+RH组),采用TUNEL检测海马神经元的凋亡,Western Blot检测Bcl-2、Bax蛋白的表达。结果:与NC组比较,HG组海马神经元凋亡率显著升高(P<0.01);RI+RH组凋亡率显著降低(P<0.01)。与HG组比较,RI组、RH组、RI+RH组凋亡率均显著降低(P<0.01)。与RI组比较,RI+RH组凋亡率显著降低(P<0.01)。与NC组比较,HG组Bcl-2蛋白表达、Bcl-2/Bax显著降低(P<0.01,P<0.05);RI组、RH组Bcl-2蛋白表达显著升高(P<0.01),RI+RH组Bcl-2蛋白表达显著降低(P<0.01),RI组、RH组、RI+RH组Bax蛋白表达显著降低(P<0.01),RI组、RH组、RI+RH组Bcl-2/Bax显著升高(P<0.01)。与HG组比较,RI组、RH组Bcl-2蛋白表达显著升高(P<0.01),RI+RH组Bcl-2蛋白表达显著降低(P<0.01),RI组、RH组、RI+RH组Bax蛋白表达显著降低(P<0.01),RI组、RH组、RI+RH组Bcl-2/Bax显著升高(P<0.01)。与RI组比较,RI+RH组Bcl-2、Bax蛋白表达显著降低(P<0.01),Bcl-2/Bax显著升高(P<0.01)。结论:红景天苷联合胰岛素能够抑制高糖培养海马神经元凋亡,其作用优于单用胰岛素及红景天苷的效果,抗凋亡机制可能与上调细胞内Bcl-2/Bax的比值有关。 Objective: To explore the effects of rhodioside combine with insulin on Bcl-2 and Bax expression and the apoptosis of hippocampal neurons cultured in high glucose. Methods: Hippocampus were obtained from 15 newborn 24 h SD rats and then primary cultured. Accordingto the purpose of the study, hippocampal neurons were divided into 5 groups: normal control group(NC group), high glucose group(HG group), insulin group(RI group), rhodioside group(RH group) and insulin combain with rhodioside(RI+RH group). The apoptosis of neurons was detected with TUNEL. And the expressions of Bcl-2 and Bax were detected by using Western Blot. Results: Compared with the NC group, the apoptosis rate of hippocampal neurons in HG group was increased significantly(P〈0.01); the apoptosis rate of hippocampal neurons in RI+RH group was decreased significantly(P〈0.01). Compared with the HG group, the apoptosis rate of hippocampal neurons in RI group RH group and RI+RH group was decreased significantly(P〈0.01). Compared with the RI group, the apoptosis rate of hippocampal neurons in RI+RH group was decreased significantly(P〈0.01). Compared with the NC group, the expression of Bcl-2 and Bcl-2/Bax of HG group was decreased significantly(P〈0.01, P〈0.05), and the expression of Bcl-2 of RI group and RH group was increased significantly(P〈0.01), and the expression of Bcl-2 of RI+RH group was decreased significantly(P〈0.01), and the expression of Bax of RI group RH group and RI+RH group was decreased significantly(P〈0.01), and the Bcl-2/Bax of RI group, RH group and RI+RH group was increased significantly(P〈0.01). Compared with the HG group, the expression of Bcl-2 of RI group and RH group was increased significantly(P〈0.01), and the expression of Bcl-2 of RI+RH group was decreased significantly(P〈0.01), and the expression of Bax of RI group, RH group and RI+RH group was decreased significantly(P〈0.01), and the Bcl-2/Bax of RI group RH group and RI+RH group was increased significantly(P〈0.01). Compared with the RI group, the expression of Bcl-2 and Bax of RI+RH group was decreased significantly(P〈0.01), and the Bcl-2/Bax was increased significantly(P〈0.01). Conclusion: Rhodioside combine withinsulin could reduce the apoptosis of neurons cultured in high glucose, and its effect is better than that of single insulin, which may be achieved by increasing the ratio of Bcl-2/Bax in the cell.
