摘要
试验建立了同时测定饲料中恩诺沙星和喹乙醇药物含量的超高效液相色谱-串联质谱(UPLC/MS/MS)分析方法。采用80%乙腈(含1%甲酸)作为样品的提取溶液,经HLB固相萃取柱净化后上机测定。在正离子模式下通过两对离子对进行定性和定量分析。结果显示,恩诺沙星和喹乙醇在5-100μg/l范围内线性关系良好,相关系数r均大于0.998;空白饲料在0.1、0.2、0.5mg/kg的添加浓度上的回收率均在70%-90%之间;批内、批间相对偏差(RSD)均小于20%;本方法对饲料中恩诺沙星和喹乙醇的定量限为0.1mg/kg(S/N〉10),检测限为0.05mg/kg(S/N〉3)。
A method based on ultra performance liquid chromatography-tandem mass spectrometry has been developed for the determination of enrofloxacin and olaquindox in feeds. The analytes were ex- tracted with 80% acetonitrile containing 1% formic acid, and were cleaned by the HLB cartridge. The purified extracts were analysed by UPLC-MS/MS under the mode of positive electrospray ionization. The results indicated that in the range of 5 to 100μg/l, the correlations of enrofloxacin and olaquin- dox were greater than 0.998. The average recoveries of enrofloxacin and olaquindox at three concentra- tion levels of 0.1, 0.2 mg/kg and 0.5 mg/kg ranged from 70% to 90%. The RSD were less than 20%. The limits of quantity(S/N〉10), and limits of detection(S/N〉3) by the present method for the enrofloxacin and olaquindox were found to be 0.1 mg/kg and 0.05 mg/kg respectively.
