响应面法优化短梗五加总皂苷的超声提取工艺 被引量:1

Optimization of ultrasonic-assisted extraction of total saponin from Acanthopanax sessiliflorus using response surface methodology

摘要 以短梗五加茎为原料,采用超声波辅助提取短梗五加中总皂苷。研究料液比、乙醇体积分数、超声时间、超声功率四个因素对短梗五加中总皂苷提取效果的影响,在单因素实验的基础上,采用Box-Behnken Design实验,以总皂苷得率为响应值,建立回归模型并由此得出四个因素最优提取工艺组合为:料液比1∶50,乙醇体积分数68%,超声时间41 min,超声功率350 W,此条件下短梗五加总皂苷的得率为1.577%。实验真实值与模型预测值1.586%基本一致,表明响应面中心组合法合理可行,可以优化提取短梗五加总皂苷的工艺。 The root stem of Acanthopanax sessiliflorus( Rupr. et Maxim.)Seem was used as the raw material, ultrasonic assisted extraction of total saponins from Acanthopanax sessiliflorus.The effect of material-to-liquid ratio, ethanol concentration, ultrasonic time, ultrasonic power on the extraction result of the total saponin of Acanthopanax sessiliflorus was studied. On the basis of single factor experiment, Box- Behnken Design was applied to build a regression model, with the yield rate of total saponin as response value. Establish regression model and according to this,the optimal extraction combination was follows:material-to-liquid ratio ]:50, ethanol concentration 68% ,ultrasonic time 4! min, ultrasonic power 350 W. In this condition,the yield of the total saponin was 1.577%.The true value of the experiment accords with the predictive value of the model was 1.586% ,show that response surface center combination method is reasonable and feasible, which can optimize the extraction process of total saponins from Acanthopanax sessiliflorus.
