高危型人乳头瘤病毒单一类型感染和多重感染对宫颈癌中细胞周期蛋白、抗凋亡蛋白表达量的影响 被引量:8

摘要 目的 研究高危型人乳头瘤病毒(hr-HPV)单一类型感染和多重感染对宫颈癌中细胞周期蛋白、抗凋亡蛋白表达量的影响。方法 选择2010年1月~2016年1月在西北妇女儿童医院接受手术切除治疗的宫颈癌患者118例,收集宫颈癌组织,测定HPV的类型并分为hr-HPV阳性和hr-HPV阴性、hr-HPV多重感染和hr-HPV单一感染。检测宫颈癌组织中细胞周期蛋白Cyclin B1、Cyclin D1、CDK1、CDK6以及抗凋亡蛋白Bcl-2、Bcl-xl、Survivin的表达量。结果 hr-HPV阳性92例、hr-HPV阴性26例,hr-HPV多重感染37例、hr-HPV单一感染55例。hr-HPV阳性宫颈癌组织中Cyclin B1、Cyclin D1、CDK1、CDK6、Bcl-2、Bcl-xl、Survivin的蛋白含量显著高于hr-HPV阴性宫颈癌组织,差异有统计学意义(P〈0.05)。hr-HPV多重感染宫颈癌组织中CyclinB1、CyclinD1、CDK1、CDK6、Bcl-2、Bcl-xl、Survivin的蛋白含量显著高于hr-HPV单一感染宫颈癌组织,差异有统计学意义(P〈0.05)。结论 hr-HPV多重感染能够通过增加宫颈癌组织中细胞周期蛋白、抗凋亡蛋白表达量的方式来加速细胞周期进程、抑制细胞凋亡,进而促进癌细胞的增殖。 Objective To study the effect of single type and multiple infection of high risk human papillomavirus (hr- HPV) on the expression of cell cycle protein and anti-apoptotie protein in cervical cancer. Methods 118 cervical cancer patients received surgical resection in Northwest Women and Children's Hospital during January 2010 to January 2016 were enrolled and cervical cancer tissues were collected. HPV types were determined and divided into hr-HPV positive and hr-HPV negative, hr-HPV multiple infection and hr-HPV single infection. The expressions of CyclinB1, CyclinD1, CDK1, CDK6 and anti apoptotic protein Bel-2, Bcl-xl and Survivin in cervical cancer tissues were detected. Results Hr-HPV positive was 92 cases, hr-HPV negative was 26 cases, hr-HPV multiple infection was 37 cases, hrHPV single infection was 55 cases. The protein contents of CyclinB1, CyclinD1, CDK1, CDK6, Bel-2, Bcl-xl and Survivin in hr-HPV positive cervical carcinoma tissues were significantly higher than those in hr-HPV negative cervical carcinoma tissues, the differences were statistically significant (P 〈 0.05). The protein contents of CyclinB1, CyclinDl, CDK1, CDK6, Bcl-2, Bcl-xl and Survivin in multiple hr-HPV infection cervical cancer tissues were significantly higher than those in single infection cervical carcinoma, the differences were statistically significant (P 〈 0.05). Conclusion Multiple infection of hr-HPV can accelerate the progression of cell cycle and inhibit apoptosis of cell through upregulating the expression of cell cycle proteins and anti-apoptosis protein in cervical carcinoma, thereby promoting cancer cell proliferation.
