产黄酮甘草内生真菌YF-A的发酵工艺优化 被引量:5

Optimization of fermentation technology for flavonoids-producing endophytic fungus YF-A of Licorice

摘要 以产黄酮甘草内生真菌YF-A为研究对象,对其发酵产黄酮的工艺进行了优化。首先确定了该菌株合适的碳、氮源种类以及发酵周期,然后选取碳源浓度、氮源浓度、发酵起始p H值、发酵温度等4个发酵影响因素进行了单因素及正交试验。结果表明,菌株合适的碳氮源为葡萄糖、蛋白胨,发酵周期为4 d。氮源浓度和发酵温度对菌株YF-A的黄酮产量影响极显著(P<0.01),其次为碳源浓度的影响(P<0.05),发酵起始p H值的影响很小。得到的优化发酵工艺条件为:培养基中葡萄糖25 g/L、蛋白胨1.5 g/L,p H值调节至7.0,接种后在120 r/min、32℃下发酵4 d。在此条件下,得到的黄酮产量为143.4μg/m L,比优化前的黄酮产量(108.5μg/m L)提高32.2%,检测到甘草苷、异甘草苷、甘草素3种甘草黄酮成分,含量分别为25.8、0.45、22.55μg/m L。 The flavonoids-producing endophytic fungus YF-A was studied in this paper, and its fermentation technology for flavonoids production was optimized. The type of carbon source and nitrogen source, and the fermentation cycle were firstly determined. The single factor experiments and orthogonal test were developed using carbon source, nitrogen source, initial pH value and fermentation temperature as influencing factors. The results showed that the appropriate carbon source and nitrogen source were glucose and peptone, and the fermentation cycle was determined as 4 d. The effects of nitrogen source and fermentation temperature on the flavonoids production were highly significant(P〈0.01), secondly the effect of carbon source(P〈0.05), and the effect of initial pH was very small. The optimum fermentation technology obtained as follows: the glucose concentration in medium was 25 g/L, and the peptone concentration was 1.5 g/L, the initial pH value was 7.0, the inoculum was incubated under 120 r/min and 32 ℃ for 4 d. Under the optimized conditions, the total flavonoids production was 143.4 μg/mL, which was 32.2% higher than the flavonoids production(108.5 μg/mL) before optimization. Three licoflavone components liquiritin, isoliquiritin and glycyrrhizin were detected in the fermentation broth, with the contents of 25.8, 0.45, 22.55 μg/L, separately.
