RP-HPLC法同时测定复方苦木消炎片中5种成分

摘要 目的建立RP-HPLC法同时测定复方苦木消炎片(穿心莲和苦木)中5种成分的含有量。方法该药物50%乙醇提取液的分析采用Agilent Eclipse Plus C18色谱柱(250 mm×4.6 mm,5μm);流动相乙腈(A)-0.1%磷酸(B),梯度洗脱;体积流量1.0 m L/min;柱温25℃;检测波长254 nm(0~26 min,4-甲氧基-5-羟基铁屎米酮)、225 nm(26~40 min,穿心莲内酯)、205 nm(40~55 min,新穿心莲内酯、去氧穿心莲内酯、脱水穿心莲内酯)。结果4-甲氧基-5-羟基铁屎米酮、穿心莲内酯、新穿心莲内酯、去氧穿心莲内酯、脱水穿心莲内酯分别在0.055~1.378、0.099~2.470、0.045~1.115、0.041~1.030、0.073~1.813μg范围内呈良好的线性关系,平均加样回收率分别为98.6%、99.1%、97.1%、101.0%、97.8%,RSD(n=6)分别为1.3%、0.98%、1.3%、1.6%、1.7%。结论该方法简便快捷,可用于复方苦木消炎片的质量控制。 AIM To establish an RP-HPLC method for the simultaneous content determination of five constituents in Compound Kumu Xiaoyan Tablets( an anti-inflammatory agent containing Andrographis Herba and Picrasmae Ramulus et Folium). METHODS The analysis of 50% ethanol extract of this drug was performed on a 25 ℃thermostatic Agilent Eclipse Plus C18column( 250 mm × 4. 6 mm,5 μm),with the mobile phase comprising of acetonitrile( A)-0. 01% phosphate acid( B) flowing at 1 m L / min in a gradient elution manner,the detection wavelengths were set at 254 nm( 0- 26 min) for 4-methoxy-5-hydroxycanthin-6-one,225 nm( 26- 40 min) for andrographolide,and 205 nm( 40- 55 min) for neoandrographolide,deoxy-andrographolide and dehydroandrographolide. RESULTS 4-Methoxy-5-hydroxycanthin-6-one,andrographolide,neoandrographolide,deoxy-andrographolide and dehydroandrographolide showed good linear relationships within the ranges of 0. 055- 1. 378 μg,0. 099- 2. 470 μg,0. 045- 1. 115 μg,0. 041- 1. 030 μg and 0. 073- 1. 813 μg,whose average recoveries were98. 6%,99. 1%,97. 1%,101. 0% and 97. 8% with the RSDs( n = 6) of 1. 3%,0. 98%,1. 3%,1. 6% and1. 7%,respectively. CONCLUSION This simple method can be used for the rapid quality control of Compound Kumu Xiaoyan Tablets.
