油茶根际土壤解磷菌的筛选、鉴定及培养条件 被引量:8

Isolation,Identification and Culture Condition of Phosphate-solubilizing Bacteria Derived from Camellia Rhizosphere Soil

摘要 从油茶根际土壤中分离筛选出7株溶磷细菌,根据透明圈法和钼锑抗比色法综合分析,最终得出菌株4-Y-06溶磷活性最强。通过菌落形态特征、生理生化特征、16S r DNA序列和系统发育分析等研究,初步鉴定菌株4-Y-06为嗜气芽孢杆菌。同时,研究了不同碳源、氮源、C/N、p H、以及温度等不同培养条件对菌株4-Y-06溶磷效果的影响。结果表明:解磷菌4-Y-06在碳源为蔗糖、氮源为硫酸铵、C/N为40:1、p H 7.0-7.5、30℃条件下解磷效果最好。 Seven strains phosphate solubilizing bacteria were screened from the rhizosphere of camellia,and strain 4-Y-06 with the highest capacity of solubilizing phosphate was gained,by utilizing the method of transparent zone and molybdenum-antimony anti-spectrophotometric.According to morphology of the colony,physiological and biochemical properties,16 S r DNA and phylogenetic analysis,Strain 4-Y-06 was identified as Bacillus aerophilus.In addition,the ability of phosphate solubilization by strain 4-Y-06 was studied with different cultivation conditions including differenr carbon sources,nitrogen forms,C / N,p H,temperatures.The results showed that strain 4-Y-06 had the highest capacity of phosphate solubilization when sucrose was used as carbon source,ammonium sulfate was used as nitrogen form,C / N was 40∶ 1,p H was 7.0-7.5 and the temperature was 30 ℃.
