摘要
目的:探讨二甲双胍对结直肠癌细胞(SW480、HCT116)的放射敏感性的影响。方法:采用CCK-8法检测二甲双胍、X射线及二甲双胍联合X射线及AMPK抑制剂干预对SW480、HCT116细胞增殖的影响。应用流式细胞仪检测二甲双胍、X射线、二者联合等干预后SW480、HCT116细胞的凋亡情况。应用Western blotting检测二甲双胍、X射线、二者联合处理SW480、HCT116细胞后相关蛋白的表达情况。结果:CCK-8结果显示:二甲双胍对SW480、HCT116细胞的增殖有抑制作用,并呈现浓度、时间依赖;X射线对SW480、HCT116细胞也有增殖抑制作用,8Gy时对SW480、HCT116细胞的增殖抑制作用最显著(P<0.05);在二甲双胍(1 mmol/L)与X射线(8Gy)共同处理SW480、HCT116细胞,联合组对细胞的增殖抑制作用均显著高于单用组(P<0.05)。流式细胞术检测结果显示:二甲双胍(1mmol/L)与X射线(8Gy)联合诱导细胞凋亡率显著高于二者单用时(P<0.05)。Western blotting结果显示:与单用二甲双胍组(1 mmol/L)、单用X射线组(8Gy)比较,联合组的p-AMPK表达显著增加,而p-m TOR的表达显著下降。结论:二甲双胍能够增加结直肠癌细胞的放射敏感性。
AIM:To explore the effects of met- formin for the radiation sensitivity of colorectal cancer cells. METHODS:CCK-8 method was used to test the effects of metformin, X-rays and metformin combined X-ray and AMPK inhibitor intervention for the proliferations of SW480 and HCTll6 cells. Flow cytometry was used to test the apoptosis of SW480 and HCTll6 cells after treatment with metformin, X-rays and metformin combined X-ray. Western blotting was used to detect the protein expression of p-AMPK and p-mTOR of SW480 and HCTll6 cells. RESULTS:CCK-8 showed that the proliferation of SW480 and HCTll6 cells were significantly inhibi- ted by metformin in concentration- and time-depend- ent manners. X-ray ( 8Gy ) also markedly sup- pressed the proliferation of SW480 and HCTll6 cells (P 〈 0.05 ). Metformin ( 1 mmol/L) and X- rays ( 8Gy ) together ( combined-treatment group )revealed that the SW480 and HCT116 cells prolifer- ration were significantly inhibitted compared with metformin group or X-ray group ( P 〈 0.05 ). Flow cytometry detection results found that the apoptosis rate of combined-treatment group were significantly higher than metformin group or X-ray group (P 〈 0.05). Western blotting results showed that the ex- pression of p-AMPK of combined-treatment group were significantly increased compared with the 1 mmol/L metformin group or 8Gy X-rays group, but the expression of p-mTOR of the combined-treatment group decrease markedlyd. CONCLUSION: Met- formin increased the radiosensitivity of colorectal cancer cells .
