摘要
【目的】对泽泻、决明子、绞股蓝等3种中草药超微粉用量进行优化组合,探索降低鸡蛋胆固醇的中草药复方添加剂最佳配方。【方法】选取泽泻、决明子、绞股蓝超微粉用量3因素为自变量,以蛋黄胆固醇浓度为响应值,采用响应面法中Box-Behnken模型,共设计17种中草药复方添加剂配方,选取190日龄绿壳蛋鸡255羽,随机均分成17组,分别饲喂添加了17种中草药复方添加剂配方的基础日粮,测定饲喂第28天各组鸡蛋蛋黄胆固醇浓度,确定中草药复方添加剂最优配方。另取190日龄绿壳蛋蛋鸡30羽,平均分成2组,空白组饲喂基础日粮,试验组饲喂基础日粮+最优配方中草药添加剂,饲喂28d后测定蛋鸡生产性能、鸡蛋品质、蛋黄胆固醇和甘油三酯浓度、血清生化指标、免疫指数等,评估最优中草药复方添加剂配方的饲喂效果。【结果】响应面优化得到的降低鸡蛋胆固醇中草药复方添加剂的最佳配比为:泽泻超微粉用量695.49 mg/只,决明子超微粉用量893.14 mg/只,绞股蓝超微粉用量334.13mg/只,该条件下饲喂28d后测定的鸡蛋胆固醇浓度为2.48mmol/L。饲喂效果验证试验结果表明,与空白组相比,试验组的料蛋比降低了33.64%,产蛋率提高了11.73%,差异均达显著水平(P<0.05);蛋黄胆固醇和甘油三酯浓度分别降低了69.90%和41.78%(P<0.05),血清胆固醇、甘油三酯、低密度脂蛋白胆固醇浓度分别下降了52.90%,55.23%和51.85%,并显著提高了蛋鸡的免疫能力(P<0.05)。【结论】响应面法能够用于中草药复方添加剂配方优化;用得到的中草药复方添加剂最优配方饲喂蛋鸡,能显著降低鸡蛋胆固醇浓度,可在实际生产中推广应用。
【Objective】This study aimed to optimize the contents of superfine comminution of Alisma orientale,Semen cassia and Gynostemma pentaphyllumin compound Chinese herb additives to reduce the content of cholesterol in egg.【Method】Using contents of Alisma orientale,Semen cassia and Gynostemma pentaphyllum powder as independent variables and egg yolk cholesterol concentration as response variable,Box-Behnken model of response surface method was used with 17 compound additive formulas for Chinese herbal medicine.A total of 255 190-day old green-shell chicken were chosen and divided into 17 groups.The optimal recipe of Chinese herbal additives was determined by detecting the content of cholesterol in the egg after 28 days.Another 30 chicken were divided randomly into two groups,the blank group was only fed with based diet,and the experimental group were fed with basal diet and optimal recipe of Chinese herbal additives.After 28 days,the performance,egg quality,egg yolk cholesterol levels,serum biochemical parameters,and immune index were measured to assess the effect of Chinese herb additives.【Result】The best Chinese herb additives composition of Alisma orientale,Semen cassia and Gynostemma pentaphyllum were 695.49 mg,893.14 mg,and 334.13 mg per chicken,respectively.The egg cholesterol concentration was 2.48 mmol/L under these composition conditions after 28 days.Compared with control group,the feed-egg ratio was dropped by 33.64%(P0.05),the egg-laying rate was increased by 11.73%(P0.05),and the egg yolk cholesterol and triglyceride levels were dropped by 69.90%and 41.78%(P0.05),respectively.The immune index was significantly increased(P0.05).Serum cholesterol,triglycerides,and low-density lipoprotein-cholesterol concentrations were decreased by 52.90%,55.23%,and51.85%.【Conclusion】Response surface methodology could be adopted for the optimization of Chinese herb additive formulation,and the obtained formulation lowered cholesterol eggs significantly and suitable for application.
