长链非编码RNA H19对胰腺癌细胞增殖、侵袭及转移的影响 被引量:1

摘要 目的:探讨下调长链非编码RNA H19的表达对胰腺癌SW1990细胞增殖、侵袭转移能力的影响。方法:从4种胰腺癌细胞Patu8988、SW1990、Bxpc3、Panc-1中筛选出H19高表达株与低表达株。对高表达H19的SW1990细胞株瞬时转染H19 siRNA(实验组),同时转染阴性siRNA作为对照组。采用qRT-PCR检测转染效率;CCK-8法检测细胞增殖活性;平板克隆法检测细胞克隆形成能力;Transwell试验检测细胞迁移和侵袭的变化;蛋白质印迹法检测细胞增殖和凋亡相关蛋白的表达。结果:与对照组相比,实验组H19 mRNA表达明显降低(P<0.05),细胞相对增殖率无变化(P>0.05),但细胞克隆形成数、细胞侵袭及转移能力明显提高,ZEB1 mRNA表达增加,但mir-200家族的表达无明显变化(P>0.05),ZEB1、Snail及PCNA蛋白表达增加,Bcl/Bax比值升高(P均<0.05)。结论:下调H19表达可提高细胞克隆形成能力,促进细胞侵袭转移。 Objective:To investigate the effects of long-coding RNA H19 on proliferation,migration and invasion of pancreatic cancer cells.Methods:We first detected the expression of H19 in Patu8988, SW1990,Bxpc3 and Panc-1 pancreatic cancer cells with real-time quantitative PCR (qRT-PCR).The SW1990 and Bxpc3 cells had higher H19 expression while Patu8988 and Panc-1 had lower H19 expression. The H19 siRNAs were transfected into pancreatic cancer cells SW1990 in vitro.The mRNA expression of ZEB1 and mir-200 were analyzed with qRT-PCR.The effects of H19 on proliferation were evaluated by CCK-8 and colon formation assays.The expression of cell proliferation-and EMT-related proteins were de-tected with Western blotting.Results:Compared with the control group,SW1990 cells transfected with H19 siRNAs revealed a significant lower level of H19 and lower the ability of cell colony formation.Howev-er,there was no significant change on cell proliferation rate;the expressional levels of ZEB1,Snail and PC-NA were up-regulated as well as the ratio of Bcl-2 to Bax;the mRNA level of ZEB1 was up-regulated while the expression of miR-200 showed no significant change.Conclusion:Down-regulation of H19 promotes cell proliferation,migration and invasion in pancreatic cancer cells SW1990.
