摘要
目的:探讨癌相关蛋白EZH2和钙黏蛋白(E-Cadherin)在子宫内膜异位症大鼠模型异位灶中的表达及意义。方法:根据大鼠自体移植方法构建大鼠子宫内膜异位症模型为实验组(n=15),选取未造模的正常大鼠为对照组(n=10)。应用免疫组织化学法(Elivision法)和Western-blot法检测实验组异位灶内及对照组子宫内膜中EZH2和E-Cadherin的表达情况,并分析两者的相关性。结果:免疫组化检测EZH2在实验组异位灶和对照组子宫内膜组织中阳性表达的平均光密度值分别为(43.10±1.52)和(32.70±1.64),而E-Cadherin在实验组异位灶和对照组子宫内膜组织中阳性表达的平均光密度值分别为(15.70±1.64)和(23.30±1.64),差异均有统计学意义(P<0.05)。Western-blot法检测实验组异位灶中EZH2表达量(195.26)明显高于对照组(85.74),而E-Cadherin表达量(24.67)明显低于对照组(92.50),差异均有统计学意义(P<0.05)。实验组EZH2与E-Cadherin的表达呈负相关(r=-0.9626,P<0.05)。结论:子宫内膜异位症病灶中EZH2高表达都伴随着ECadherin的低表达,且两者呈明显的负相关,EZH2可能通过抑制E-Cadherin的表达促进子宫内膜异位症的发生及发展。
Objective:To study expression and significance of the cancer associated protein EZH2 and mucins E-Cadherin in the ectopic foci of endometriosis rats model. Methods:According to the method of autologous transplantation in rats, rats with endometriosis model was constructed for the experimental group ( n = 15), select not building normal rats for blank group( n = 10). Immune histochemical method( Elivision method) and Western Blot method were used to detect the expressions of EZH2 and E-Cadherin in the experimental ectopic foci tissue and in the endometrium tissue of blank group,the correlation between the two indexes were analyzed. Results:By the immunohistochemical detection,the mean positive expression density of EZH2 protein in ectopic foci of experimental group and endometrial tissue of blank group was (43. 10 ±1.52 ) and (32. 70 ±1.64), respectively; and the mean positive expression density of E-Cadherin protein in ectopic foci of experimental group and endometrial tissue of blank group was( 15. 70± 1. 64 )and (23. 30 ±1.64 ), respectively, the differences between the two groups were statistically significant( P 〈0. 05). By Western Blot method,the EZH2 protein expression in the experimental ectopic foci(195.26) was higher than that of blank group(85. 74), E-Cadherin protein expression in the experimental ectopic foci(24. 67) was lower than that of blank group(92.50),the differences were statistically significant( P 〈 0. 05). In the experimental group, EZH2 protein expression was negatively correlated with the expression of E-Cadherin protein(r= -0. 9626, P 〈0. 05). Conclusions:The high EZH2 expression in endometriosis lesions is associated with E-Cadherin lower expression,and the two indexes are negative correlated with each other, EZH2 may promotes the occurrence and development of endometriosis by inhibiting the expression of E-Cadherin.
