摘要
目的观察芪葵颗粒对糖尿病肾病大鼠肾脏中转化生长因子β_1(TGF-β_1)及单核细胞趋化蛋白1(MCP-1)的影响,初步探讨其作用机制。方法本研究采用高脂高糖饲料喂养+单侧肾结扎+腹腔注射2%链脲佐菌素(STZ)造成糖尿病肾病大鼠模型,成模后将其随机分为模型组,厄贝沙坦组,芪葵颗粒高、中、低剂量组,另设假手术组,每组10只,连续灌胃给药8周。血糖仪测定大鼠血糖值,酶联免疫吸附测定(ELISA)法检测血清中白细胞介素6(IL-6)的含量,免疫组化法检测肾组织中MCP-1、TGF-β_1的表达。结果与模型组比较,芪葵颗粒高、中剂量组大鼠血糖显著降低,差异有统计学意义(P<0.05);与实验前比较,芪葵颗粒高、中、低剂量组大鼠血糖均显著降低(P<0.01或P<0.05);与模型组比较,芪葵颗粒高、中、低剂量组大鼠血清中IL-6含量明显降低(P<0.01),肾组织中MCP-1、TGF-β_1的表达明显减少(P<0.01)。结论芪葵颗粒对糖尿病肾病大鼠肾组织具有较好的保护作用,其作用机制可能与其降低血糖、减少IL-6和抑制MCP-1、TGF-β_1的表达有关。
Objective To observe the effects of Qikui Granules on the expression of transforming growth factor β1 (TGF-β1) and monocyte chemotactic protein 1 (MCP-1) in kidney of rats with diabetic nephropathy, and to evaluate the mechanism. Methods This experiment adopted the high-sugar-high-fat diet + unilateral renal ligation + intraperitoneal injection of 2% STZ to establish rat models with diabetic nephropathy, then the rats were randomly divided into model group, Irbesartan group, Qikui Granules of high, medium, low dose group, and sham group, with 10 rats in each group. Continuous intragastric administration of 8 weeks was taken. The values of blood glucose in rats were measured by blood glucose meter. The contents of serum interleukin 6 (IL-6) were determined by enzyme linked immunosorbent as- say (ELISA). The expression of MCP-1 and TGF-β1 in kidney was evaluated by immunohistochemistry method. Results Compared with model group, the levels of blood glucose in Qikui Granules of high, medium dose groups were decreased (P 〈 0.05). The levels of blood glucose in Qikui Granules of high, medium, low dose groups were significantly lower than those before experiment (P 〈 0.01 or P 〈 0.05). Compared with model group, the contents of IL-6 in Qikui Granules of high, medium, low dose groups were significantly decreased (P 〈 0.01), the expression of MCP-1 and TGF-β1 in kidney of Qikui Granules of high, medium, low dose groups was significantly reduced (P 〈 0.01). Conclu- sion Qikui Granules have better protective effects for the renal tissue of rats with diabetic nephropathy. Its mechanism may be related to the decrease of blood glucose and IL-6, the inhibition of the expression of MCP-1 and TGF-β1.
