摘要
目的研究芎麻汤药效部位对硝酸甘油诱导的偏头痛大鼠血浆中核因子κB(NF-κB)含量及三叉神经颈髓复合体(TCC)内NF-κB阳性表达的影响,初步探讨芎麻汤药效部位的作用机制。方法 36只雄性SD大鼠随机分为6组,分别为空白对照组,模型空白组,芎麻汤药效部位低、中、高剂量组,琥珀酸舒马普坦片阳性对照组;采用皮下注射硝酸甘油(10mg·kg^(-1))复制偏头痛模型,给药后取颈外静脉血采用酶联免疫吸附测定法(ELISA法)检测血浆中NF-κB的含量,心脏灌注后取大鼠TCC部位采用免疫组织化学检测NF-κB的阳性表达。结果偏头痛模型大鼠外周血中NF-κB浓度显著升高,与空白对照组比较,差异具有统计学意义(P<0.01),模型复制成功;芎麻汤药效部位能有效降低偏头痛大鼠TCC上NF-κB免疫阳性染色光密度值,与模型组比较,差异具有统计学意义(P<0.05)。结论芎麻汤药效部位能下调硝酸甘油型偏头痛大鼠TCC内NF-κB表达,从而改善偏头痛发作时三叉神经血管系统激活后引起的神经源性炎症反应,达到缓解偏头痛症状。
Objective To study the bioactive parts of Xiongma decoction to NF - κB in plasma and trigeminocervical complex on animal model of migraine with nitroglycerin. Methods 36 SD rats were randomly divided into 6 groups: blank group, model group, the bioactive parts of Xiongma decoction of low, middle and high dose group, Sumatriptan group. By giving hypodermic in- jection of lOmgokg - 1 nitroglycerin, copied the migraine model, to observe the content of CGRP on plasma with ELISA method, and the expression of NF - KB on TCC with immunohistochemistry. Results NF - KB on TCC of the model group could effectively increase, compared with the blank group, the difference was significant(P 〈0.01 ) , shows the model copy success; The bioac- tive parts of Xiongma decoction could effectively decrease the content of NF - κB in TCC, compared with the model group, the difference was significant( P 〈 0.05 ). Conclusion The bioactive parts of Xiongma decoction regulated the expression of NF - κB, so as to improve the neurogenic inflammation reaction caused by activation of the trigeminovascular system after the migraine attack, relieve migraine symptoms.
