摘要
[目的]本文旨在研究Th表位肽和聚肌胞苷酸[Poly(I:C)]2种免疫增强剂对猪O型口蹄疫合成肽抗原细胞免疫和体液免疫的增强作用。[方法]将上述2种免疫增强剂分别与O型口蹄疫病毒(FMDV)合成肽抗原混合乳化,分组免疫Balb/c小鼠,分别在免疫后7、14、21和28 d检测小鼠血清中的VP1抗体、IL-4和IFN-γ水平,以及脾脏中T淋巴细胞转化率及脾脏组织形态变化。[结果]免疫后14 d Th表位肽疫苗组和Poly(I:C)疫苗组血清VP1抗体水平显著高于普通疫苗组(P<0.05),免疫后28 d Th表位肽疫苗组显著高于Poly(I:C)疫苗组和普通疫苗组(P<0.05)。细胞因子检测结果显示:免疫后14、21 d Th表位肽与Poly(I:C)疫苗组IL-4含量显著高于普通疫苗组(P<0.05),免疫后14 d Th表位肽疫苗组IFN-γ含量明显高于其他组(P<0.05),免疫后21 d Th表位肽与Poly(I:C)疫苗组IFN-γ含量显著高于普通疫苗组(P<0.05)。Th表位肽与Poly(I:C)均可使试验小鼠脾脏白髓区增大,着色加深,动脉周围淋巴鞘胀大,Poly(I:C)疫苗组淋巴细胞增殖系数显著高于其他各试验组(P<0.05)。[结论]Th表位肽和Poly(I:C)对猪O型口蹄疫合成肽抗原均有显著免疫增强作用,而Th表位肽可延长体液免疫持续时间。
[ Objectives] This paper was aimed to study the immunity enhancement of swine foot-and-mouth disease virus(FMDV) type O synthetic peptide vaccine by Th epitope peptides and poly inosine-cytidine [ Poly(I:C) ]. [ Methods ] Mix these two immunopotentiators with the synthetic peptides of swine FMDV type O and emulsify it separately. Then, inject the Balb/c mice subcutaneously. Explore the change of VP 1 antibody, IL-4 and IFN-γ cytokines, T lymphocyte proliferation and morphological observation in spleen at the day of 7,14,21 and 28 after immunization. [ Results] The indirect ELISA shows that the levels of VP1 antibody of Th epitope peptides group and Poly(I:C) vaccine group are significantly higher than vaccine group at 14 day after immunization(P〈0.05). The levels of VP 1 antibody of and Poly(I:C) vaccine group are significantly higher than vaccine group at 14 day after immunization(P〈 0.05). The Th epitope peptides group expresses significantly higher antibody level than other groups at 28 day. The cytokine analysis shows that Th epitope peptides and Poly( I : C) can significantly increase serum level of IL-4 than the other groups at 14,21 day(P〈 0.05). Th epitope peptides group expresses higher significantly serum level of IFN-',/than the other groups at 14 day(P〈0.05), and the serum levels of IFN-γ of Th epitope peptides group and Poly(I:C) vaccine group are higher than vaccine group. Th epitope peptides and Poly(I: C)can both enlarge the white pulp area of experimental animals' spleen, deepen the dyeing and swell the peripheral arterial lymphatic sheath. Poly ( I : C) vaccine increases significantly lymphocyte proliferation coefficient ( P 〈 0.05 ). [ Conclusions ] Th epitope peptides and Poly ( I : C ) can both enhance the immune response of the vaccine, while the Th epitope peptides have a stronger ability to extend the time of humoral immune response.
