摘要
目的:初步研究不同生长环境和采收期对问荆药材中异槲皮苷的影响,并建立定性和定量的分析方法。方法:采用薄层色谱法,对问荆药材中异槲皮苷进行定性分析;采用HPLC法对问荆药材中异槲皮苷进行含量测定,Z O R B A X SB-C1 8(4.6 m m×2 5 0 m m,5μm)为色谱柱;流动相:乙腈-0.1%甲酸(1 5∶8 5),流速为1.0 m L·m in-1;检测波长为354 nm;进样量为10μL。结果:采收期对问荆药材的薄层色谱和异槲皮苷含量均有较大的影响;薄层色谱斑点清晰,分离度良好;异槲皮苷在0.41~95.13μg·m L-1范围内与峰面积呈良好线性,r=0.9999,平均回收率为9 7.2%,R SD=1.3%。结论:每年6~8月是问荆药材的最佳采收期;所建立的方法简便,灵敏,重现性良好,可作为问荆药材的质量控制方法。
Objective: Study on different growth environment and harvest time on the quality of Equisetum arvens L . To establish methods for identification and determination of isoquercitrin in Equisetum arvens L. Methods: TLC was used to identify isoquercitrin in Equisetum arvens L, and the content was determine by HPLC, Agilent ZORBAX SB-C18(4. 6 mm × 250 mm,5 μm) column was used. Acetonitrile and 0. 1% Formic acid water-solution were used as mobile phase of liner elution, and the volume flow was at 1.0mL· min-1 , Uhraviolent determination wavelength was 354nm; and injection volume was 10μL Results: Different growth environment and harvest time influence obviously TLC and the content of isoquercitrin. TLC spots were clear, and sparated nicely. The linear range of isoquercitrin was 0. 41-95.13μg·mL-1. The average recovery in Equisetum arvens L was 97. 2%, RSD was 1.3%. Conclusion: June and August are the best harvest time. The method is believable for identitication and determination of isoquercitrin in Equisetum arvens L. with its simplicity, sensibility, repeatability.
