Shh信号通路相关基因表达调控甲状腺功能减退的机制 被引量:3

摘要 目的探讨甲状腺功能减退(甲减)大鼠海马组织中Shh信号通路信号肽Shh、膜受体Patched-1和核转录因子Gli-1的表达及甲减对脑发育及功能调控影响的分子机制。方法 12只SPF级健康Wistar大鼠按照随机字数表法分为对照组和甲减组,每组6只,甲减组经腹腔注射丙基硫氧嘧啶建模,对照组经腹腔注射生理盐水,测定两组三碘甲状腺原氨酸(T3)、T4及促甲状腺激素(TSH)水平,对比两组Shh、Patched-1和Gli-1蛋白水平及mRNA表达。结果与对照组比较,甲减组大鼠血清T3、T4水平明显降低,TSH水平明显升高(P<0.05)。甲减组Shh,Patched-1及Gli-1蛋白及mRNA表达相比对照组均明显降低(P<0.05)。结论甲减脑组织Shh、Patched-1和Gli-1蛋白水平及mRNA表达明显降低,导致甲状腺激素生物学效应降低,为甲状腺功能减退的临床治疗打下坚实基础。
