养肝益水颗粒对自发性高血压大鼠肾脏保护作用的影响 被引量:4

摘要 目的观察养肝益水颗粒对自发性高血压大鼠(SHR)血压、尿微量白蛋白(尿MA)、肾脏组织中内皮素、一氧化氮、血管性假性血友病因子水平的影响。方法将SHR大鼠随机分为中药组(养肝益水颗粒)、西药组(卡托普利组)、模型组,每组8只。另外选取Wistar-Kyoto大鼠8只作为正常对照组。灌胃6周后检测其血压、尿MA、肾脏组织中内皮素(ET-1)、一氧化氮(NO)、血管性假性血友病因子(vWF)的表达。结果中药组能够降低SHR大鼠血压进行性增高的趋势;治疗后尿MA低于模型组(P<0.01);中药组肾脏组织中ET-1、vWF的表达较模型组显著下降,有统计学意义(P<0.01);而NO水平较模型组显著升高(P<0.01)。结论中药养肝益水颗粒可以减弱SHR大鼠血压增高趋势,持久降低血压,减少SHR大鼠尿MA的水平,降低肾组织中ET-1、vWF的表达,升高NO的水平,从而起到保护高血压肾损害的作用。 Objective To investigate the effect of Yanggan Yishui granule(YYG)on the levels of blood pressure,urinary microalbuminuria(MA)in urine,kidney tissue endothelin-1(ET-1),nitricoxide(NO),von Willebrand factor(vWF)in spontaneously hypertensive rats(SHR),and its mechanism of renal protection of SHR.Methods SHR were randomly divided into YYG group,captopril group,model group,8 each group.Eight Wistar-Kyoto rats were as normal control group.After 6 weeks,the blood pressure,urinary MA,ET-1 in the kidney tissue,the expression of nitric oxide(NO)and vWF were tested.Results The blood pressure trend of progressive increase reduced in YYG group.Urinary MA was lower than that in model group after treatment(P .01).Compared with model group,the expressions of kidney tissue ET-1,vWF decreased significantly in YYG group(P〈0.01).NO concentration significantly increased compared with model group(P〈0.01).Conclusion YYG has renal protective effect,which can reduce increased blood pressure,lower blood pressure,reduce the levels of urine MA,decrease the expressions of ET-1 and vWF in renal tissue,increase the levels of NO in SHR.
