摘要
目的:探索二甲基亚砜(dimethyl sulfoxide,DMSO)对骨髓瘤细胞系RPMI8226细胞的影响。方法:利用细胞增殖-毒性检测试剂盒(Cell Counting Kit-8,CCK-8)检测不同浓度DMSO作用于RPMI8226细胞不同时间后对细胞的增殖抑制作用。利用吖啶橙染(AO)色在倒置荧光显微镜下观察细胞的形态变化。结果:DMSO浓度≤0.6%时各处理组与对照组细胞存活率无影响,当DMSO≥0.8%时,随着浓度的增加以及作用时间的延长,DMSO对RPMI8226细胞的增殖抑制作用逐渐增强,呈浓度-时间依赖性(P<0.05),且在荧光显微镜下可观察到凋亡细胞。结论:DMSO浓度≤0.6%时,对RPMI8226细胞基本无影响,当浓度≥0.8%,DMSO具有抑制细胞增殖促进细胞凋亡的作用。
Objective To study the effects of DMSO on proliferation of multiple myeloma cell line RPMI8226. Method Cell Counting Kit-8( CCK-8) was used to detect the effects of diferent concentrations of DMSO on proliferation of RPMI8226 cell lines for different time. Acridine orange( AO) was used to stain the cells. The apoptotic morphology of cells was examined using acridine orange( AO) stains. Results When DMSO≤0. 6%,the cell viability rate of treatment group had no difference with control group. when DMSO≥0. 8%,the proliferation inhibition of DMSO on RPMI8226 cells gradually strengthened with increasing of DMSO concentrations and extension of time. Apoptosis was confirmed by nuclear condensation,membrane blebbing,and formation of round apoptotic bodies. Conclusion When DMSO≤0. 6%,the DMSO have no effect on RPMI8226 cells almostly. when DMSO≥0. 8%,the DMSO can inhibit the proliferation of RPMI8226 cells and induce apoptotice.
