摘要
采用酶解结合物理法对雨生红球藻的破壁工艺进行了研究,以破壁孢子率为评价指标,通过正交试验筛选酶解条件,对比试验进行验证。试验结果表明,雨生红球藻的最佳破壁工艺为:选用复合纤维素酶对雨生红球藻进行酶解处理,酶添加量0.5%,酶解温度45℃,酶解时间5 h,酶解后结合高压均质法进行破壁,均质压力40 MPa,均质2次。在此工艺条件下,雨生红球藻的破壁孢子率较原工艺提高18.07%。酶解结合高压均质法用于雨生红球藻粉破壁具有效率高、成本低的优点,适用于工业化生产。
Tostudythe cell wall-breakingprocess byenzyme combined with physical method for Haematococcus pluvialis. Methods:Cell wall-breaking rate was as the evaluation index.Orthogonal test method was used to concern enzymolysis conditions, and verified by contrast experiment. Results: The enzymolysis conditions identified as: composite cellulose enzyme, adding amount of 0.5%,enzyme solution temperature 45 ℃, enzymolysis time 5 h,homogeneous pressure 40 Mpa, homogeneous 2 times. Compared with the original method, cell wall-breaking rate of new method increased 18.07%.Conclusion: Enzyme combined with physical method is high efficiency, low cost for breaking Haematococcus pluvialiscell wall, and is suitable for industrialized production.
