金铁锁愈伤组织色素提取条件研究

摘要 金铁锁是我国特有的珍稀濒危药用植物,常用于治疗风湿、外伤出血、跌打损伤和胃脘冷痛等。本试验以金铁锁紫红色的愈伤组织为实验材料,先采用单因素试验,分别考查不同提取条件(提取剂、体积分数、时间、温度、p H和料液比)对色素提取效果的影响,再设计正交试验对色素提取的四个条件(体积分数、料液比、时间和温度)进行优化;并且使用紫外-可见分光光度法和高效液相色谱法,通过比较标准品(矢车菊-3-O-葡萄糖苷)及色素提取液的吸收峰及出峰时间,对提取的金铁锁色素进行初步鉴定。结果表明,影响色素提取的因素次序是:料液比>时间>温度>体积分数。金铁锁色素的适宜提取条件为提取剂料液比1∶10(g∶m L)、提取时间20 h、提取温度30℃和2%盐酸-甲醇。并初步鉴定了金铁锁愈伤组织色素的主要色素成分为矢车菊素。 Psammosilene tunicoides is a rare and endangered medicinal plant in China. It is commonly used in treating rheumatism, traumatic hemorrhage, injuries and gastric abscess and so on. In this study, red callus of Psammosilene tunicoides was used to extract pigment. The effect of different conditions such as solvent, volume fraction, the length of extraction, extraction temperature, pH and material-liquid ratio on extraction rate of pigments were investigated. Based on one-factor-at-a-time experiments, the orthogonal experiment was designed to optimize the extracting conditions of pigment. The ultraviolet-visible spectrophotometry and high performance liquid chromatography were adopted to analyze the extraction. Psammosilene tunicoides extracting conditions were based on comparing the absorption peaks and appearance time of standard substance (cyanidin-3-O-glucoside) and extracted pigment. The results showed that the order of contributing factors were : material-liquid ratio 〉 extraction time 〉 extraction temperature 〉 volume fraction. And the optimum values of extraction parameters were determined by orthogonal array : material-liquid ratio 1 : 10, extraction time 20 h, extraction temperature 30~C with 2% HCI ethanol solution. The main pigment of Psammosilene tunicoides callus was identified as cyanidin.
