摘要
研究向日葵盘果胶(SFHP)分子量分布对其体外抗氧活性的影响。制备SFHP,通过超滤技术将其分为〉100 k D、10-100 k D、〈10 k D三个级分。利用DPPH·、·OH及O-2·三种体外抗氧化模型对各级分的抗氧化活性进行研究。结果表明,对于·OH,分子量分布为10-100 k D级分清除率最高,优于粗提总果胶,又优于分子量分布〈10 k D及〉100 k D的果胶;对于DPPH·,分子量分布〈10 k D的级分作用最强,当果胶浓度为6.4 g/L时,对DPPH·的清除率达到最大92.78%;对于O-2·,粗提果胶及各级分的清除作用均很弱,三个级分果胶相比,〉100 k D级分的清除作用较强,果胶浓度为0.2 g/L时,得到最大清除率为28.68%。在所研究的浓度范围内,向日葵盘果胶各级分对DPPH·、·OH及O-2·的清除率均随着果胶浓度变化呈现极值行为。研究表明分子量分布对SFHP的抗氧化活性影响显著。
The effect of the molecular weight distribution of sunflower head pectin on its antioxidant activity in vitro was investigated. The molecular weight of sunflower head pectin(SFHP)was fractionated by MWCO. The antioxidant activity of different molecular weight of SFHP were investigated by the antioxidant model of DPPH ·,· OH,O- 2· in vitro. For · OH,the highest clearance rate was the pectin which molecular weight was among 10 ~ 100 k D. For DPPH · scavenging ability,pectin with molecular weight less than 10 k D showed the highest clearance rate,and the maximum scavenging ability of 92.78% was obtained at pectin concentration of 6.4 g/L. SFHP showed poorly antioxidant activity for O- 2·. The pectin with molecular weight more than 100 k D showed stronger antioxidant activity,and the maximum scavenging ability of 28.68% was obtained at the concentration of 0.2 g/L. In the range of pectin concentration in the study,the scavenging ability of DPPH ·,· OH,O- 2· showed extreme values with pectin concentration. Therefore,the molecular distribution had a significant effect on antioxidant activity of SFHP.
