摘要
目的探讨七氟醚分娩镇痛对缺氧缺血性脑损伤(HIBD)新生大鼠脑组织中c-fos表达的影响。方法将120只发情期成年雄性和雌性Sprague-Dawley大鼠合笼饲养(雌性∶雄性=3∶1),选择其中妊娠雌性大鼠72只随机分为自然分娩组、模型组(前列腺素诱导胎鼠HIBD)和七氟醚干预组(大鼠吸入体积分数2.5%七氟醚),每组24只。自然分娩组、模型组和七氟醚干预组大鼠分别分娩新生大鼠141、133、140只,处死新生大鼠并取脑组织。采用实时定量聚合酶链反应检测新生大鼠脑组织中c-fos mRNA表达水平,Western blotting法检测新生大鼠脑组织中c-fos蛋白表达水平。结果自然分娩组、模型组和七氟醚干预组新生大鼠脑组织中c-fos mRNA表达水平分别为0.77±0.31、5.03±0.93和2.00±0.70,c-fos蛋白表达水平分别为1.00±0.26、7.40±1.30和3.93±1.15;模型组和七氟醚干预组新生大鼠脑组织中c-fos mRNA及其蛋白表达水平显著高于自然分娩组(P<0.05),七氟醚干预组新生大鼠脑组织中c-fos mRNA及其蛋白表达水平显著低于模型组(P<0.05)。结论七氟醚可能通过下调c-fos表达而发挥对新生大鼠HIBD的保护作用。
Objective To study the effect of sevoflurane labor analgesia on the expression of c-fos in brain tissue of neonatal rats with hypoxic-ischemic brain damage( HIBD). Methods A total of 120 adult male and female Sprague-Dawley rats were fed in the same cage( female∶male = 3∶1). The 72 pregnant rats were selected and randomly divided into spontaneous delivery group,model group( the fetal rats with HIBD were induced by prostaglandin) and sevoflurane intervention group( the rats were interfered with 2. 5% sevoflurane by inhalation),24 rats in each group. There were 141,133 and 140 neonate rats in spontaneous delivery group,model group and sevoflurane intervention group respectively. The neonatal rats were sacrificed,and the brain tissues were collected. The level of c-fos mRNA in brain tissue of neonatal rats was detected by quantitative real-time polymerase chain reaction detection. The level of c-fos protein in brain tissue of neonatal rats was detected by Western blotting. Results The level of c-fos mRNA in brain tissue of neonatal rats in spontaneous delivery group,model group and sevoflurane intervention group was 0. 77 ± 0. 31,5. 03 ± 0. 93 and 2. 00 ± 0. 70 respectively; and the level of c-fos protein was1. 00 ± 0. 26,7. 40 ± 1. 30 and 3. 93 ± 1. 15 respectively. The levels of c-fos mRNA and c-fos protein in brain tissue of neonatal rats in model group and sevoflurane intervention group were significantly higher than those in spontaneous delivery group( P〈0. 05),and the levels of c-fos mRNA and c-fos protein in brain tissue of neonatal rats in sevoflurane intervention group were significantly lower than those in model group( P〈0. 05). Conclusion Sevoflurane may play a protective effect on HIBD by down-regulating the expression of c-fos in neonatal rats.
