摘要
对名贵中药材冬虫夏草及其伪品建立稳定可靠的双向电泳鉴定方法。正品冬虫夏草中共同表达的蛋白质斑点为88个,伪品稳定差异表达的蛋白质斑点为21个,提示可作为下一步冬虫夏草鉴定的指标性组分。进一步质谱分析与NCBI数据库比对表明:6种蛋白仅来自于菌数据库,9种蛋白仅来自于昆虫数据库,12种蛋白仅来自于全库,3种蛋白昆虫和菌的数据库均能检索,1种蛋白仅来自于菌数据库和全库,提示下一步可进行冬虫夏草指标性蛋白质功能研究,为冬虫夏草有效成分研究奠定基础。
The study is aimed to establish an efficient two-dimensional gel electrophoresis protocol for proteomic studying of Ophiocordyceps sinensis. Using two-dimensional electrophoresis, a proteomic analysis of O. sinensis from different areas and other Cordyceps fungi were compared. 88 kinds of proteins in O. sinensis were found. Compared with other Cordyceps fungi, 21 kinds of characteristic proteins spots in O. sinensis were found. These indicated that the 21 kinds of proteins were able to be used to authenticate O. sinensis from other Cordyceps fungi. Then further analysis of the proteins Mass Spectra Database of NCBI, 6 kinds of proteins came from the fungi database, 6 kinds of proteins from insect database, 12 kinds of proteins from the total database, 3 kinds of proteins were found both in insect database and fungi database, 1 kinds of proteins was found in the fungi database and total database. These indicate that further study is needed for some function protein.
