摘要
目的研究中药益心康对感染柯萨奇B_3病毒(Coxsackie virus B,CVB_3)新生乳鼠体外心肌细胞凋亡相关基因caspase-3 mRNA和颗粒酶B、穿孔素(PFP)表达的影响,探讨中药益心康治疗病毒性心肌炎的作用机制。方法原代培养SD大鼠乳鼠心肌细胞,建立体外感染CVB_3的心肌细胞模型,将心肌细胞分为4组:正常对照组、病毒组、中药益心康组及利巴韦林组。用RT-PCR法检测凋亡相关因子caspase-3 mRNA的表达,ELISA法检测颗粒酶B、PFP的表达。结果中药益心康组caspase-3 mRNA、PFP、颗粒酶B的相对表达量明显低于病毒组(P<0.01)和利巴韦林组(P<0.05)。结论中药益心康能降低大鼠心肌细胞caspase-3 mRNA、颗粒酶B、PFP表达,从而抑制CVB_3感染大鼠心肌细胞凋亡。
Objective To study the effect of Yixin Kang on the expression of cardiomyocytes apoptosis related gene caspase-3 mRNA and granzyme B,pore-forming protein( PFP) in vitro in neonatal rat with infection of CVB_3 virus,and discuss the action mechanism of Yixin Kang in the treatment of viral myocarditis. Methods In order to establish an in vitro model of CVB_3 virus infection,the primary culture of SD rats was established in vitro. The neonatal rat cardiomyocytes were divided into 4 groups: control group,virus group,Chinese herbal medicine Yixin Kang group and Ribaviri group. RT-PCR method were used to detected the expression of apoptosis related factor caspase-3 mRNA,and the expression of granzyme B and PFP by ELISA. Results The expression of caspase-3 mRNA,granzyme B and PFP of Yixin Kang group were significantly lower than that of the virus group( P 〈 0. 01) and ribavirin group( P 〈0. 05). Conclusion Lixin Kang can decrease the expression of caspase-3 mRNA,granzyme B and PFP in cardiac muscle cells of rat so as to inhibit the cardiomyocytes apoptosis in CVB_3 virus infected rats.
