摘要
目的探讨创伤性休克对兔肾组织不同时相中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肾损伤分子-1(Kim-1)与热休克蛋白70(HSP70)表达的影响。方法健康新西兰大白兔30只,随机分为对照组、0 h模型组、6 h模型组、12 h模型组和24 h模型组,每组6只。通过酶联免疫吸附法(ELISA)、蛋白质印迹法(Western-blot)与实时荧光定量聚合酶链反应(Real-time PCR)测定各组兔血液、尿液及肾组织中NGAL、Kim-1、HSP70的含量与mRNA的表达变化。结果与对照组比较,模型组各时相血液及尿液中NGAL、Kim-1及HSP70含量均升高,6 h开始增加,24 h时有所恢复,但仍为高表达,差异有统计学意义(P<0.05)。与对照组比较,模型组兔肾组织在不同时相NGAL、Kim-1及HSP70表达量均显著升高,在6 h时升高最明显,差异均有统计学意义(P<0.05)。结论 NGAL、Kim-1、HSP70与创伤性休克导致肾损伤的发生和发展相关。监测血液、尿液及肾组织中NGAL、Kim-1及HSP70的变化情况,有助于对创伤性休克导致肾损伤的判断和治疗。
Objective To explore the expression of neutrophil gelatinase associated lipocal(NGAL),kidney injury molecule-1(Kim-1),heat shock protein 70(HSP70) in rabbit at different time points after traumatic shock were studied to explore their possible role.Methods A total of 30 healthy New Zealand white rabbits were randomly divided into control group,0 hour model group,6 hours model group,12 hours model group and 24 hours model group.ELISA,Western blot and Real-time PCR were used to detect the change and expression of NGAL,Kim-1,HSP70 and their mRNA in blood,urine and nephridial tissue.Results Compared with control group,the expression of NGAL,Kim-1 and HSP70 in blood and urine 6hours increased after the traumatic shock,and then recovered at 24 hours,was still in the high expression(P 0.05).The expression of NGAL,Kim-1 and HSP70 tissues and mRNA expression in nephridial tissue of modeling groups of rabbits were also significantly elevated,and reached the highest in 6 hours(P 0.05).Conclusion Kim-1 and HSP70 are related to the occurrence and development NGAL of kidney injury in traumatic shock;monitoring the change of NGAL,Kim- 1,and HSP70 in blood,urine and nephridial tissue will help to the carry out judgment and treatment of kidney injury in traumatic shock.
