摘要
目的探讨葛根饮料对小鼠酒精性肝损伤的保护作用。方法将实验小鼠随机分为模型组、空白对照组、阳性对照组、高浓度、中浓度、低浓度组6组,分别用体积分数50%的乙醇灌胃,造成小鼠酒精性肝损伤,灌胃量为12m L/kg·BW,同时,除模型组外,高、中、低三浓度组分别灌胃实验室配制的1/2倍、1倍和2倍浓度的葛根饮料,阳性对照组灌胃RU-21药,空白组灌胃生理盐水,每天灌胃2次,每次间隔12h。实验四周后,取小鼠肝脏组织,测定各组小鼠肝匀浆丙二醛(MDA)浓度、谷胱甘肽过氧化物酶(GSH-Px)、甘油三酯(TG)含量和血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)活力,评估葛根饮料对小鼠酒精性肝损伤的治疗作用。结果与模型组相比,3个浓度组小鼠肝匀浆GSH-Px含量明显升高(P<0.01),TG含量明显下降(P<0.05),高浓度组、阳性对照组小鼠血清ALT含量下降(P<0.05)。结论葛根饮料对小鼠酒精性肝损伤具有一定的肝脏保护作用。
OBJECTIVE The purpose of the present study was to investigate the effect of pueraria lobata beverage on alcohol-induced liver injury in mice. METHODS Divided mice into 6 groups,each group was intragastric administration with 50% ethanol( 12 m L·kg-1). At the same time,high,medium and low concentration groups were lavaged 1 /2,1 and 2 times content of puerarin beverages,positive group was lavaged RU-21 medicine,blank group was lavaged saline. They were lavaged 2 times per day. After experiment,the technique was performed to measure the malondialdehyde( MDA) concentration,glutathione peroxidase( GSH-Px),triglycerides( TG) levels and serum aspertate aminotransferase( AST),as well as alanine aminotransferase( ALT). RESULTS The results showed that,compared with alcohol-induced group,pretreatment lavaged with low,normal,high content pueraria lobata beverage groups,homogenate gsh-px levels increased significantly( P〈0. 01),TG content significantly decreased( P〈0. 05),high concentration group,positive control group mice serum ALT levels decreased( P〈0. 05). CONCLUSION Pueraria lobata beverage can protect alcoholic liver injury.
