骨痛灵方对肺癌骨癌痛模型小鼠的疼痛行为学以及脊髓MCP-1和NGF的影响 被引量:8

Effects of Gutongling Prescription on the pain behavior in mice model of lung cancer with bone metastasis and MCP-1 and NGF in the spinal cord

摘要 目的观察骨痛灵方对肺癌骨癌痛模型小鼠脊髓单核细胞趋化因子(MCP-1)和神经生长因子(NGF)表达的影响,探讨骨痛灵方治疗骨癌痛的潜在作用机制。方法在C57小鼠左后肢胫骨骨髓腔注射Lewis肺癌细胞建立骨癌痛模型。50只小鼠随机分为假手术组、模型组、中药组、西药组和中西药组,每组10只。中药组小鼠以骨痛灵方煎剂灌胃,西药组予以唑来膦酸腹腔注射,中西药组则两药联合运用。用Von Frey纤维及热痛测试仪分别于造模前,造模后第6(7)天、13(14)、20(21)天观察小鼠机械性缩足反射阈值(PWMT)和热痛觉缩足潜伏期(PWTL)的变化,酶联免疫吸附反应(ELISA)检测脊髓MCP-1和NGF蛋白的表达情况。结果3周后,3个治疗组的PWMT和PWTL均明显高于模型组(均P<0.05),中西药组的PWMT比中药组和西药组均增高,差异有统计学意义(均P<0.05)。与模型组比较,中药组、西药组和中西药组的MCP-1蛋白表达均明显降低(均P<0.05);NGF则均显著升高(均P<0.05)。结论骨痛灵方可能通过减少脊髓MCP-1蛋白的表达,抑制中枢敏化,同时促进NGF蛋白的表达,修复中枢敏化中受损的神经元而发挥止痛作用。 Objective To observe the effects of Gutongling Prescription on the expression of monocyte chemoattractant protein-1( MCP-1) and nerve growth factor( NGF) in the spinal cord of mice with bone metastasis of lung cancer model,and discuss its potential mechanism on the treatment of bone cancer pain. Methods The mice model of lung cancer with bone metastasis was established by the injection of Lewis lung cancer cells into the bone marrow cavity of left leg tibia. 50 mice were randomly divided into five groups: sham-operation group,model group,Chinese medicine group,western medicine group,Chinese and western medicine combination group,10 in each mice. The Chinese medicine group was treated with decoction of Gutongling Prescription for intragastric administration,the western medicine group was treated with zoledronic acid for intraperitoneal injection,the combination group was treated with Gutongling Prescription and zoledronic acid.Before modeling and 6( 7),13( 14),20( 21) days after modeling,the changes of paw withdrawal mechanical threshold( PWMT) and paw withdrawal thermal latency( PWTL) were observed by Von Frey hairs and heat stimulation test. The protein expressions of MCP-1 and NGF in the spinal cord were detected by enzyme linked immunosorbent assay( ELISA). Results 3 weeks after modeling,the PWMT and PWTL of the three treatment groups were obviously higher than those of the model group( P〈 0. 05),the PWMT of the combination group was higher than that of the Chinese medicine group and western medicine group,with statistically significant difference( P 〈0. 05). Compared with the model group,the protein expression of MCP-1 in all the three treatment groups was obviously decreased( P 〈0. 05),and the NGF expression was significantly increased( P 〈0. 05). Conclusion Gutongling Prescription can suppress the central sensitization by decreasing the protein expression of MCP-1 in the spinal cord,and repair the damaged neurons by promoting the protein expression of NGF,and consequently exert the analgesic effect.
