摘要
近年来研究表明,生物钟的紊乱可影响药物的代谢或毒性,进而影响疾病的发生发展。细胞色素P450酶(CYPs)是重要的药物代谢酶,参与临床上70%~80%常用药物的代谢。研究发现许多CYPs显示明显的昼夜节律变化,这一现象受到生物钟的调控,并且关于其调控机制的研究在近些年也获得了一定的进展。本综述拟在介绍生物钟作用的基础上,从直接途径及生物钟控制基因、核受体等间接途径阐述生物钟如何对CYPs的昼夜节律变化进行调控,从而影响药物的代谢和解毒。
In recent years,studies show that the disorder of the biological clock can affect the metabolism or toxicity of drugs,and then affect the occurrence and development of the disease. Cytochrome P450 enzymes(CYPs) are the important drug metabolism enzymes involved in the metabolism of 70% ~ 80% commonly used drugs. Many CYPs display obvious circadian rhythmic changes,and this phenomenon is controlled by the biological clock. Recently progress has been achieved in the regulation of biological clock on CYPs. Based on the introduction of biological clock function,we elaborated how biological clock regulates the circadian rhythm of CYPs through the direct pathway or indirect way such as biological clock controlled gene and nuclear receptors,thus affecting drug metabolism and detoxification.
