摘要
目的研究大蒜素对人喉癌Hep-2细胞周期与细胞凋亡的影响。方法实验分为空白对照组、大蒜素25μg/m L组、大蒜素50μg/m L组、大蒜素100μg/m L组和顺铂40μg/m L组,取对数生长期人喉癌Hep-2细胞,每组分别给予相应干预24 h后,采用MTT法测定细胞增殖抑制率,通过流式细胞术检测细胞周期、观察细胞凋亡状况并计算凋亡率,RTPCR法检测bcl-2 mRNA和bax mRNA表达并计算bax/bcl-2表达比值,Western blotting法检测Caspase-3蛋白表达情况。结果与空白对照组比较,大蒜素50μg/m L组、大蒜素100μg/m L组和顺铂40μg/m L组人喉癌Hep-2细胞增殖抑制率显著升高,细胞周期G0/G1期显著增长、G2/M期显著缩短,细胞凋亡率显著升高,bcl-2 mRNA表达显著下调而bax mRNA显著上调,bax/bcl-2表达比值显著升高,Caspase-3蛋白表达显著上调,差异均有统计学意义(P均<0.05)。大蒜素100μg/m L组细胞增殖抑制率、细胞凋亡率和Caspase-3蛋白表达量均显著高于顺铂40μg/m L组(P均<0.05)。结论大蒜素具有阻滞人喉癌Hep-2细胞周期进程和促进其凋亡的作用,机制可能与其能够调节凋亡相关基因和蛋白表达有关。
Objective It is to observe the influence of allicin on human laryngeal cancer Hep-2 cell cycle and apoptosis.Methods The experiment was divided into blank control group,allicin 25 μg / m L group,allicin 50 μg / m L group,allicin 100μg /m L group,cisplatin 40 μg /m L group,corresponding intervention for logarithmic growth phase of human laryngeal carcinoma Hep-2 cells in each group was given for 24 hours. Then the inhibition rate of cell proliferation was determined by MTT assay; the cell cycle was detected by flow cytometry,and the apoptosis rate and apoptosis rate were observed; the expression of bcl-2 mRNA and bax mRNA was detected by RT-PCR,and the bax / bcl-2 ratio was calculated; the expression of caspase-3protein was detected by Western blotting method. Results Compared with blank control group,the cellular growth inhibition rate of allicin 50,100 μg / m L groups and Cisplatin 40 μg / m L group were significantly increased; the cell cycle was arrested at G0/ G1 phase,and G2/ M period were significantly shortened; the apoptosis rate was significantly increased; the expression of bcl-2 mRNA was significantly decreased,while the expression of bax mRNA was significantly increased,and the ratio of bax /bcl-2 was significantly increased; the expression of caspase-3 protein were significantly increased; all of the difference above were significant( all P〈0. 05). The cell proliferation inhibition rate,apoptosis rate and the expression of Caspase-3 protein in allicin 100 μg / m L group were significantly higher than those of cisplatin 40 μg / m L group. Conclusion Allicin can block human laryngeal cancer Hep-2 cell cycle progression and induce apoptosis,which perhaps related to its effects of altering the expression of apoptosis-related genes and proteins.
