加减驻景方含药血清对CoCl_2诱导ARPE-19细胞表达VEGF及HIF-1α的影响 被引量:4

摘要 目的探讨加减驻景方含药血清对二氯化钴(CoCl_2)诱导后人视网膜色素上皮细胞(ARPE-19细胞系)中血管内皮生长因子(VEGF)及缺氧诱导因子-1α(HIF-1α)蛋白表达的影响。方法 1.体外培养ARPE-19细胞,取对数生长良好的细胞用于实验,分为正常组,缺氧模型组,阳性对照组、含药血清组,并设立不同时间点。2.酶联免疫吸附试验(ELISA)观察CoCl_2诱导后不同时间、各组细胞上清液中HIF-1α的表达。3.蛋白质印迹法(Western blot)观察CoCl_2诱导后不同时间、各组细胞中VEGF、HIF-1α的表达情况。结果缺氧损伤后可诱导ARPE-19细胞中VEGF、HIF-1α蛋白的表达增加;与正常组比较,缺氧模型组VEGF及HIF-1α的表达高于正常组,差异具有统计学意义(P<0.05);含药血清组与模型组比较,VEGF及HIF-1α的表达低于模型组,差异具有统计学意义(P<0.05)。结论加减驻景方含药血清对缺氧损伤后ARPE-19细胞中VEGF、HIF-1α蛋白的表达有抑制作用。 OBJECTIVE To explore effects of serum with modified Zhujing prescription on expression of vascular endothelium growth factor (VEGF) and hypoxia-inducible factor1α (HIF-1α) protein in cobalt chloride induced ARPE-19 cells. METHODS 1. The ARPE-19 cells cultured in vitro and cells in well logarithmic growth were chosen to be assigned randomly into normal group, hypoxia group, positive control groups and medicated serum group. Time interventions were set for observation. 2. Effects on expression of HIF-1α in supernate at different time point were assessed by ELISA. 3. The expression of VEGF and HIF-1α protein in ARPE-19 cell under hypoxia were detected by Western Blot. RESULTS Expression of VEGF and HIF-1α increased after hypoxic injury, in addition, they were higher than counterparts of normal group. The difference was statistically significant (P〈0.05). While com- pared with model group, expression of VEGF and HIF-1α in medicated serum group were higher statistically (P〈 0.05). CONCLUSIONS Medicated serum with modified Zhujing prescription inhibited the expression of VEGF and HIF-1α in ARPE-19 cells under hypoxia injury.
