摘要
目的建立了饮料中苯甲酸、山梨酸钾两种防腐剂同时检测的高效液相色谱法。方法饮料经前处理后,由C18反相色谱柱进行分离,用流动相:甲醇和0.02mol/ml的乙酸铵溶液以5∶95为比例洗脱,在流速1.0ml/min,波长230nm下进行检测,进样量为10μl,以保留时间定性,峰面积定量。结果一次进样分析仅需18min。该方法的线性范围为0~400μg/ml。苯甲酸的相关系数r=0.998 8,山梨酸钾的相关系数r=0.999 8,检出限0.001 7μg/ml,回收率99.65%,相对偏差1.3%。结论该法准确、快速、简便,可推广应用。
Objective A HPLC method for the determina- tion of benzoic acid, potassium sorbate in beverage was established. Methods The sample was extracted and the components were separated on a C18 reversed phase column eluated with a mobile phase of methyl alcohol and ammonium acetate (5:95). At the wave length of 230 nm, the 10 μl sample was detected by High Per- formance Liquid Chromatography.The flow velocity was 1.0 ml/min. The method was qualified by the ratio between the retention time of chroma- tographic peak of the samples and standard substance, which means qualified the location of the chromatograph- ic peak. And to quantify it through the area of chromatographic peak is proportional to the concentration. Re- suits Analysis of one injection only need 18 min. Linearity of the calibration curve wasin the range from 0 to 400 μg/ml. The correlation index of benzoic acid and potassium sorbate were 0. 998 8, 0. 999 8 respectively. The average recovery rate was 99. 65% and relative standard deviation ( n = 5 ) was 1.3%, while the concen- tration limit was 0. 001 7 μg/ml. Conclusion Thismethod is accurate, fast and simple, and is worth to be pop- ularized and applied.
