摘要
目的研究癌胚抗原相关细胞黏附分子1(carcinoembryonic antigenrelated cell adhesion molecule 1,CEACAM1)和血管内皮生长因子(vascular endothelial growth factors,VEGF)在涎腺黏液表皮样癌(mucoepidermoid carcinoma,MEC)组织中的表达及对血管生成的影响。方法应用免疫组织化学链霉菌抗生物素-过氧化物酶连接(streptavidin-perosidase,SP)法检测70例MEC、20例正常涎腺组织中CEACAM1、VEGF的表达情况,在光镜下计数70例MEC组织中CD105标记的微血管密度(microvessel density,MVD)。结果 70例MEC组织中CEACAM1蛋白阳性表达率明显低于正常涎腺组(34.3%vs 70.0%,P<0.05),VEGF蛋白阳性表达率明显高于正常涎腺组(71.4%vs 10.0%,P<0.05)。在MEC组织中病理分级Ⅲ级者CEACAM1表达阳性率低于病理分级Ⅰ+Ⅱ级者(P<0.05),TNM分期Ⅲ+Ⅳ期者CEACAM1表达阳性率低于TNM分期Ⅰ+Ⅱ期者(P<0.05),有淋巴结转移者CEACAM1表达阳性率低于无淋巴结转移者(P<0.05);而病理分级Ⅲ级者VEGF表达阳性率和MVD值高于病理分级Ⅰ+Ⅱ级者(P<0.05),TNM分期Ⅲ+Ⅳ期者VEGF表达阳性率和MVD值高于TNM分期Ⅰ+Ⅱ期者(P<0.05),有淋巴结转移者VEGF表达阳性率和MVD值高于无淋巴结转移者(P<0.05)。结论 MEC组织中CEACAM1表达下调可能促进了VEGF的表达,从而参与MEC组织当中血管的生成,促进其侵袭和转移。
Objective To investigate the relationship of carcinoembryonic antigen related cell adhesion molecule 1 (CEACAM1) and vascular endothelial growth factors (VEGF) expression with angiogenesis in mucoepidermoid carcinoma (MEC) of salivary glands. Methods The expression of CEACAM] and VEGF was detected with immunohistochemcal method streptavidin-perosidase (SP) in 70 cases of MEC tissues, and 20 cases of normal salivary gland, the microvessel,density(MVD) was recorded under optical microscope. Results The positive expressions of CEACAM1 protein (34.3% vs 70.0%) was lower in MEC group than those in normal salivary gland group(P〈0.05), the positive expressions of VEGF protein (71.4% vs 10.0%) was higher in MEC group than those in normal salivary gland group(P〈0.05). In MEC group,the positive expressions of CEACAM1 protein in pathological grade level Ⅲ patients were lower than those in pathological grade level Ⅰ + Ⅱ patients(P〈0.05) ,the positive expressions of CEACAM1 protein in TNM stageⅢ + Ⅳ patients were lower than those in TNM stage Ⅰ + Ⅱ patients(P〈0.05) ,the positive expressions of CEACAM1 protein in patients with lymph node metastasis were lower than that in patients without lymph node metastasis (P〈 0.05). The positive expressions of VEGF protein and MVD in pathological grade level Ⅲ patients were higher than those in pathological grade level Ⅰ + Ⅱ patients(P〈0.05), the positive expressions of VEGF protein and MVD in TNM stage Ⅲ + Ⅳ patients were higher than those in TNM stage Ⅰ + Ⅱ patients(P〈0. 05), the positive expressions of VEGF protein and MVD in patients with lymph node metastasis were higher than that in patients without lymph node metastasis(P〈0.05). Conclusion Downregulationg of CEACAM1 in MEC of salivary glands may play a role in angiogenesis through increasing the expression of VEGF, thus promote tumor invasion and metastasis.
