摘要
目的 评价 HPS-1 对人肺腺癌 A549 细胞凋亡蛋白 Caspase-3 、 8 、 9 以及 Bax 、 Bcl-2mRNA 表达的影响,探究 HPS-1 抗肺肿瘤可能机制。方法 人肺腺癌 A549 细胞经低、中、高剂量HPS-1 处理不同时间后,噻唑蓝 ( MTT )法检测各组细胞的增殖情况,流式细胞术 ( FCM )法检测各组细胞凋亡率及细胞周期比例,比色法检测各组细胞凋亡蛋白 Caspase-3 、 8 、 9 的表达, RT-PCR 法检测各组细胞 Bax 、 Bcl-2mRNA 的表达。结果 MTT 结果显示,低、中、高剂量 HPS-1 对人肺腺癌A549 细胞的增殖具有明显抑制作用,且具有时间、剂量依赖性; FCM 法检测发现,低、中、高剂量HPS-1 对人肺腺癌 A549 细胞的凋亡具有促进作用,且与剂量呈正相关;比色法检测各组细胞发现,HPS-1 促进凋亡蛋白 Caspase-3 、 8 、 9 的上调,且 HPS-1 高剂量组凋亡蛋白上调更明显。结论 HPS-1具有抑制人肺腺癌 A549 细胞增殖、诱导其凋亡作用,且该作用可能与 HPS-1 阻滞细胞周期 G 1 期,上调凋亡蛋白 Caspase-3 、 8 、 9 及 BaxmRNA 表达,下调 Bcl-2mRNA 表达有关。
Objective To evaluate the expressions of Caspase-3, Caspase-8, Caspase-9 and Bax, Bcl-2 under the intervention of HPS-1 in A549 cells. Methods After treated by HPS-1, the viability of cell lines was detected through methylthiazolyldiphenyl-tetrazolium bromide (MTT). The apoptosis of cell lines was detected by flow cytometry (FCM). The expressions of Caspase-3, Caspase-8, Caspase-9 were measured by colorimetry. Results The results showed that different doses of HPS-1 inhibited the proliferation and promoted the apoptosis of A549 cells. And the expression levels of Caspase-3, Caspase-8, Caspase-9 proteins increased in a concentration-dependent manner in A549 cells treated with HPS-1, and these effects were significantly enhanced in A549 cells treated with high does of HPS-1. Conclusions HPS-1 has a powerful effect on inhibiting cellular proliferation and inducing cellular apoptosis of lung adenocarcinoma A549 cells, which may be associated with the increased protein proportion of Caspase-3, Caspase-8, Caspase-9 and expression of Pax mRNA, decreased expression of Bcl-2 mRNA.
